Objective To explore the relationship between the expression of PDHA 1 protein in human prostate cancer and its clinicopathological characteristics and prognosis.Methods 60 patients with prostate cancer who underwent radical prostatectomy in our hospital from January 2012 to January 2016 were randomly selected as the subjects of this study.After treatment,all enrolled patients were followed up for a period of 3 years up to January 2019.The prostate tissue samples of 60 patients with prostate cancer included in this study were resected after prostate cancer surgery,fixed with paraformaldehyde and embedded with paraffin.Immunohistochemical staining of PDHA 1 protein was performed.The expression of PDHA 1 protein in 60 patients with prostate cancer was statistically analyzed,and the results of immunohistochemical staining were observed.The positive expression rate and percentage of positive cells of PDHA 1 protein were statistically analyzed.The general data(age,smoking status)and clinical data(tumor T stage,M stage,PSA level,Gleason score and risk factor grade)of the patients were grouped as the grouping criteria,and the positive expression rate of PDHA 1 protein in different subgroups was calculated.The survival rate of patients was calculated,and KaplanMeier method was used to analyze the survival of 60 patients with prostate cancer,and the survival of patients with positive and negative PDHA 1 protein expression in prostate cancer tissues was further compared.Age,smoking,T stage,M stage,PSA level,Gleason score,risk factor classification,PDHA 1 positive expression and other variables were set as independent variables to be included in the univariate Cox regression analysis model affecting the prognosis of patients with prostate cancer.Univariate Cox regression analysis was performed on 60 patients with prostate cancer.Variables with statistically significant differences in the analysis results(P < 0.05)were included in the multivariate Cox regression analysis for multivariate Cox regression analysis affecting the prognosis of patients with prostate cancer.The independent risk factors and protective factors affecting the prognosis of prostate cancer were statistically analyzed.Results Immunohistochemical staining showed that PDHA 1 protein was mainly located in the cytoplasm of prostate cancer tissues,and the positive staining showed brown-yellow color,and was mainly expressed in the fibrous cells,basal cells and vascular smooth muscle cells of prostate cancer tissues.Among the 60 prostate cancer specimens,20 of them had positive PDHA 1 protein expression,with a composition ratio of 33.33%.The protein expression of PDHA 1 was negative in 40 patients,with a composition ratio of 66.67%.Among the 20 samples with positive PDHA 1 protein expression,the percentage of PDHA 1 protein-positive cells was 12% ~ 84 %,with a median of 36%.For patients with different tumor T stage,M stage,PSA level,Gleason score and risk factor grade,the positive expression rate of PDHA 1 protein was significantly different(P < 0.01).Statistical results showed that the lower the T stage,M stage,PSA level,Gleason score and risk factor grade,the higher the positive expression rate of PDHA 1 protein in prostate cancer tissues,and the difference was statistically significant(P < 0.01).The survival rate of 20 patients with positive PDHA 1 protein expression in prostate cancer tissues was 80.0%.The survival rate of 40 patients with negative PDHA 1 protein expression in prostate cancer tissues was 45.0%.Kaplan-Meier survival analysis showed that the median survival time of 60 patients with prostate cancer was 30 months.Median survival was 30 months in patients with positive PDHA 1 protein and 25 months in patients with negative PDHA 1 protein.The difference in survival time between the two groups was statistically significant(χ2 = 8.861,P = 0.003).Univariate Cox regression analysis showed that T stage,M stage,PSA level,Gleason score,risk factor grade and positive PDHA 1 expression were risk factors affecting the prognosis of patients with prostate cancer(P < 0.05),while age and smoking were not risk factors affecting the prognosis of patients with prostate cancer(P > 0.05).Multivariate Cox regression analysis showed that T stage,M stage,Gleason score and risk factor grade were independent risk factors affecting the prognosis of patients with prostate cancer(P < 0.05),and positive PDHA 1 expression was a protective factor affecting the prognosis of patients with prostate cancer(P < 0.05).Conclusion Positive expression of PDHA 1 protein is associated with better prognosis in patients with prostate cancer and is a protective factor for prognosis in patients with prostate cancer.
