Objective:Because of the importance of epidermal functions,a large range of the products has been developed to improve the skin functions.Mucopolysaccharide polysulphate,also termed “heparinoid”,has long been used to manage a variety of skin conditions.Our aim was finding the underlying mechanisms whereby certain products towards skin barrier damage and eczema dermatitis.By comparing the therapeutic effects of topical glucocorticoids,we explore whether heparinoid-containing product can be used in combination with glucocorticoids to enhance the efficacy and reduce its side effects.Methods:The acute barrier disruption of the mice was applyed with tape stripping.Following twice‐daily treatments with Heparinoid cream for 7 days,epidermal biophysical properties were measured using respective probe connected to an MPA5 physiology monitor(Courage+Khazaka Electronic Gmb H,Cologne,Germany).The expression levels of m RNA for epidermal antimicrobial peptides,differentiation,proliferation and lipid synthetic enzymes were determined by q PCR.Acute contact dermatitis model in mice is established by TPA,DNFB treatment establish mouse acute allergic dermatitis and atopic dermatitis models.Mice were divided into blank group,model treatment group,Hepariod cream group,clobetasol group and combination group.Record the general changes of mice after the first treament,observe the pathological analysis of HE staining,analyze the m RNA levels of barrier function markers and inflammatory factor.Results:1.Topical application of heparinoid-containing product can reduce TEWL levels in mice after skin barrier damage and accelerate permeability barrier recovery.2.Topical application of heparinoid-containing product can stimulates expression levels of m RNA for markers of epidermal structure,function and proliferation in mouse skin and reduce inflammation level in mice after skin barrier damage3.Topical application of heparinoid-containing product can reduce ear swelling and inflammatory cell infiltration in mice after TPA treatment,increase m RNA levels of barrier structure function markers,reduce m RNA expression levels of inflammatory factors,and have a stronger combination effect with glucocorticoids4.Topical application of heparinoid-containing product can reduce the inflammatory response in acute allergic dermatitis,increase the m RNA level of barrier structure function markers,reduce the m RNA expression level of inflammatory factors,and have a stronger combination effect with glucocorticoids5.Topical application of heparinoid-containing product can reduce eczema skin lesions in atopic dermatitis,reduce hyperkeratosis,thickened spines,dermal edema,and infiltration of inflammatory cells in mice,and restore m RNA levels of skin barrier structural function markers,Reduce the expression level of inflammatory factor m RNA;and long-term application of glucocorticoids in mice has more severe skin lesions,excessive keratinization of the epidermis,focal keratosis,thickening of the spinous layer,dermal atrophy,and inflammatory cell infiltration;In the group of heparin-containing ointment and late application of glucocorticoids,the eczema skin lesions were not obvious,the spinous layer was not significantly thickened,the inflammation was lightly infiltrated,and the expression levels of markers and inflammatory factors of barrier structures were restored.Conclusions:1.Topical application of heparinoid-containing product can significantly improve the epidermal permeability barrier homeostasis in mice in the damaged skin.2.Topical application of heparinoid-containing product can improve the symptoms of eczema dermatitis,improve the permeability barrier function of skin,reduce the inflammatory response.The combination with glucocorticoids can enhance their effects,and reduce the adverse reactions caused by long-term use of glucocorticoid.
