| Nowdays,nucleic acid extraction and amplification technique has become one of the most important means not only in criminal investigation,epidemic prevention,but also in clinical treatmen.At present,the extraction-amplification methods are mostly accomplished by the combination of individual parts: centrifugalization and PCR amplification.However,the current equipments for the mentioned application are bulky and complicated,which is not suitable for real-time,on-site and minimization related conditions.This study puts forward a novel structure,in which,only designated material can pass through the surface border between media.(named as "media valve" in this study).The structure is then intergrated into microfluidic chip,the chip is used to extract nucleic acid with the help of magnetic beads.Meanwhile,the study put forward a intergrated microfluidic,automatic device,which combines nucleic acid extraction and amplification together.The achievements of this study can solve recent nucleic acid related problems,such as time wasting,deleterious and so on.Based on the nucleic acid purification scheme proposed by ourselves and LAMP amplification technology(proposed by Notomi),a nucleic acid purification and on-chip amplification system were designed and completed,prototype was formed,and the experimental method based on prototype was proposed.The results showed that the device could be successfully used for genotyping of a DNA fragment when 25 u L blood sample is used.The theoretical analysis of the study can lay the foundation for the follow-up application research,such as diagnosis of pathogens,cancer and other clinical applications. |
