Objective:To observe the effect of normal saline injection at LU5 acupoint on the improvement of lung histology and lung function in rats with chronic obstructive pulmonary disease(COPD),and to analyze its mechanism from the Angle of inflammation and oxidative stress.The possible mechanism of NRF2 /ARE pathway was explored.To provide a scientific basis for clinical acupuncture treatment of COPD.Methods:1.Grouping and modeling: 24 SD rats were randomly divided into normal control group(normal group,n=8)and model experimental group(n=16).After 1 week of feeding,model experimental group was modeled(by fumigation + Lipopolysaccharide(LPS)airway infusion)for 11 weeks.From week 8,the model experimental group was divided into model control group(model group,n=8)and Chi Ze intervention group(Chi Ze group,n=8),and the model was continued.2.Intervention: Intervention was carried out immediately after the end of grouping in Week 8:in the Chi Ze group,0.1 m L of normal saline was injected into the unilateral LU5 acupoint once a day after modeling,with a direct puncture of 0.5 mm,once a day,left and right in turn,for consecutive 4 weeks.The model group made the same model every day and fixed capture,without intervention;3.Indicator observation: During the experiment,the rats were weighed and recorded every week,and the general situation was observed.After the intervention,the rats were fasted for18 h,anaesthetized,and four physiological loggers were used to detect lung function.After an abdominal section,5ml of blood was collected from the abdominal aorta.The remaining blood was drained as much as possible.The whole blood was centrifuged and the serum was collected.Open the chest,observe the trachea,lung tissue in general appearance,remove the fixed place right lung tissue pieces of ice slurry,measuring serum,lung tissue SOD(Superoxide Dismutase,SOD),Malondialdehyde,Malondialdehyde(MDA),ELISA method to measure IL-6,IL-8,Tumor Necrosis Factor alpha(TNF-α)content in serum and lung tissue.Tissue of the fixed part of the right lobe was taken and fixed in 4% paraformaldehyde,then embedded,sected and stained with HE,and pathological morphology was observed.In addition,2 pieces of soy-sized lung tissue from the fixation site of the left lung were collected and frozen at-80℃ with liquid nitrogen.The m RNA and protein expressions of Nrf2,HO-1and GSTM in the lung tissue were measured.Results:1.General situation: compared with the normal group,the body weight of rats in model group decreased(P < 0.05),and the hair color of rats in model group became yellow and dark;After the intervention of normal saline injection at LU5 acupoint,the body weight was significantly increased(P < 0.05).2.Lung function: Compared with normal group,lung ventilation function(ventilation per minute)of model group was decreased(P < 0.05),and lung elastic resistance had a downward trend;After the intervention of normal saline injection at LU5 acupoint,lung ventilation function(ventilation per minute,lung elastic resistance)was significantly increased(all P <0.05).3.Lung histomorphology: Compared with the normal group,the appearance of lung tissue and trachea in the model group showed emphysema and inflammatory cell infiltration,while the appearance and pathological morphology of lung and trachea in the Munze group were significantly improved;4.Inflammation and oxidative stress factors: compared with normal group,the contents of IL-6,IL-8 and TNF-α in serum and lung tissue in model group were significantly increased(all P < 0.01).MDA content had a tendency to increase,SOD activity in serum decreased(P <0.05),and SOD activity in lung tissue had a tendency to decrease.LU5 point injection of saline solution after the intervention,the serum IL-6,IL-8,there is a decrease in the content of TNF alpha(P < 0.01),IL-8 of lung tissue,decreased the content of TNF alpha(P < 0.01),decrease MDA content of lung tissue(P < 0.05),there is a downturn in the serum MDA content in lung tissue IL-6 there is a downturn in the serum and lung tissue SOD activity increased(P < 0.05);5.Expression of Nrf2,HO-1,GSTM gene and protein: Compared with model group,m RNA expression of Nrf2 in Chize group was significantly increased(P < 0.05),while protein expression of HO-1 was significantly decreased(P < 0.05),and both m RNA and protein of GSTM showed an upward trend.Conclusion:1.LU5 acupoint injection has a certain prevention and treatment effect on COPD model rats.2.The effect of LU5 acupoint injection on the prevention and treatment of COPD may be related to anti-oxidative stress and anti-inflammation.3.The anti-inflammatory and antioxidant effects of LU5 acupoint injection in the prevention and treatment of COPD ARE related to the regulation of Nrf2 gene and HO-1 protein in the Nrf2/ARE pathway.
