| Background: Cancer is one of the leading causes of death in the world.Cervical cancer is the fourth most common malignant tumor worldwide,and it is also a major health problem faced by women.Its incidence ranks second among female malignant tumors in my country.Due to the vigorous development of gene sequencing,GEO is playing an increasingly important role in bioinformatics analysis.These databases provide sequencing data for discovering new functional genes and analyzing the effects of these genes on prognosis.Therefore,the purpose of this study is to use comprehensive bioinformatics analysis to identify prognostic genes.In this study,we tried to determine the expression level of INHBA gene in cervical cancer cell lines,assess its prognostic value for cervical cancer,and explore the tumor-related functions and potential mechanisms of INHBA gene.However,there is no biological function of INHBA in cervical cancer.Immune cell infiltration refers to the migration of immune cells from the blood to the tissues and begin to play its role.Infiltrating immune cells can be separated from the tissues.By exerting anti-tumor effects,immune cells infiltrating tumors have a profound impact on the development of tumors and the success of anti-cancer treatments.It has been reported that immune infiltration in head and neck squamous cell carcinoma is related to INHBA,and immune infiltrating B cells affect the prognosis.However,the relationship between INHBA and immune cell infiltration in cervical cancer has not been reported yet.Purpose: To study the potential biomarkers of cervical cancer prognosis and immune cell infiltration;analyze the expression level of INHBA in cervical cancer,explore its biological role in cervical cancer and related molecular mechanisms.Method: Download the microarray data set of cervical cancer tissue(GSE9750)from GEO,and use R’s "limma" package to identify differentially expressed genes(DEGs),using R’s "colorspace","stringi","DOSE","cluster Profiler","pathview" Perform GO and KEGG enrichment analysis.The GEPIA platform was used to verify the expression level of INHBA in cervical cancer tumor tissues and normal samples and predict the prognosis.Use STRING to construct a protein interaction network(PPI).Use the "CIBERSORT" software to obtain the expression of immune infiltrating cells in cervical cancer tissue samples through the three packages "e1071","parallel" and "preprocess Core" in R.The lentivirus sh RNAINHBA was transfected into cervical cancer cells,CCK-8 method was used to detect proliferation,flow cytometry was used to detect apoptosis,and Transwell method was used to detect migration and invasion.Western Blot is used to assess changes in related protein levels.Results:1)Compared with normal cervical tissue samples,early CD4 T cells,inactive NK cells,M0 and M1 macrophages in cervical cancer samples are all upregulated;while inactive CD4 memory T cells,monocytes,and inactivated The expression of DC in cervical cancer samples was significantly reduced.2)The Hub gene(INHBA),which is highly related to the prognosis of cervical cancer,was identified.The expression level of INHBA in cervical cancer tissues was significantly increased,and the life cycle of the high-expressing INHBA was significantly shorter than that of the low-expressing group;down-regulating INHBA made cervical cancer Cell proliferation capacity is inhibited,migration capacity is reduced,and invasion capacity is delayed;down-regulation of INHBA also promotes apoptosis.3)Down-regulation of INHBA in cervical cancer may inhibit the expression of Snail,Slug and Twist by down-regulating the expression of TGF-β1,thereby up-regulating the expression of E-cadherin,and down-regulating the expression of N-cadherin and Vimentin,thereby affecting the EMT of tumor cells process.4)Knockdown of INHBA in cervical cancer may down-regulate the expression of MMP-2 and MMP-9 to inhibit metastasis.Conclusion:1)The Hub gene INHBA,which is highly correlated with the prognosis of cervical cancer and the proportion of immune cell infiltration obtained in this study,provide a new basis for the treatment and prognosis of cervical cancer.2)We initially confirmed that INHBA plays an important role in cervical cancer and is a potential oncogene.3)In cervical cancer,down-regulation of INHBA significantly inhibits proliferation,migration and invasion,and is expected to become a potential tumor molecular marker and therapeutic target in diagnosis.In cervical cancer,INHBA may promote EMT process and metastasis by regulating the activity of TGF-β signaling pathway. |
