| ObjectiveBy detecting the oral microbial community structure and single nucleotide polymorphism(SNP)of caries-affected and healthy people,the diversity of dental caries and the host-susceptibility gene SNP,and explore the relationship between the SNP and oral microbial community structure.Method1.The plaque microbes and buccal mucosa samples of 276 volunteers(133 in the caries group and 143 in the healthy group)were collected.The DNA of plaque samples was extracted and high-throughput sequencing was performed,and then the differences in the microbial community structure between the healthy group and the caries group were analyzed and compared.2.The DNA of the buccal mucosa sample was extracted,and 19 SNPs of 12 caries susceptibility genes were detected by nucleic acid flight mass spectrometry.Chi-square test and principal component analysis were used to compare the differences in genotype and allele frequency between the two groups.3.The omics analysis method was used to analyze the correlation between the host susceptibility gene SNP and the oral microbial community structure.Result1.Oral microbial sequencing results showed that: the oral health group and the dental caries group have a certain difference in the microbial community structure,which is mainly reflected in the difference in the abundance of their members;at the phylum level,the difference between the two groups is mainly reflected in the dominant phylum.The abundance of Fusobacteria,Bacteroidetes,and Actinobacteria in the caries group is higher than that in the healthy group,while the abundance of Firmicutes and Proteobacteria is reduced;at the genus level,the difference between the two groups is mainly the lower abundance of the bacteria,Prevotella-6,The relative abundance of Lachnoanaerobaculum,Kingella and Faccalibacterium in the caries group is higher than that in the healthy group.2.The caries susceptibility was not significantly different from 19 SNP sites(P>0.05).3.Correlation analysis between dental caries susceptibility gene SNP and microbial diversity showed that rs1126478,rs1800972 and rs1143627 were related to oral microbial community structure(P<0.05).And it is mainly reflected in the suspected caries-causing bacteria genus.ConclusionThere are certain differences in the oral microbial community structure between dental caries and healthy people,which is mainly reflected in the differences in the abundance of group members;the host susceptibility gene SNP is related to the oral microbial community structure,such as rs1126478,rs1800972,and rs1143627.Suspected pathogenic bacteria with lower abundance are influential. |
