Objective:To study the effects of Angelica oil on intestinal flora diversity in hypertensive rats based on 16 S rDNA technique,and to explore the possible mechanism of Chinese medicine in treating hypertensionMethod: 30 SHR rats were randomly divided into five groups(n = 6),i.e.model control group,angelica oil low,medium and high dose groups and valsartan group,meanwhile,Wistar rats of the same age were taken as normal control group(n = 6).Model control group and normal control group were given physiological saline without any drugs.The low,middle and high dose groups of Angelica volatile oil were given 1%,3%,9% Angelica volatile oil emulsion 2.0ml ·(kg · d)-1,and valsartan 8mg·(kg·d)-1 respectively.The method of administration was gavage.Medicine is given at certain time every day for 4 consecutive weeks.The dosage was adjusted with the change of body weight.After the experiment,the total DNA of rat intestinal microorganism was extracted and sequenced in V3 and V4 regions of 16 S rDNA.Result:1.Whether the valsartan or angelica oil had no significant effect on the overall richness and diversity of the intestinal flora of hypertensive rats.2.At the level of the Phylum,the abundance of the Proteobacteria in the high-dose group of Angelica sinensis was significantly lower than that in the model group(P < 0.05);the abundance of the D.faecalis in the dose group of Angelica sinensis was extremely high compared with the model group.Significantly lower(P < 0.01).3.At the level of the Class,the abundance of the denitrifying strain of the Angelica sinensis volatile oil group was significantly lower than that of the model group(P < 0.01);the abundance of the high-dose group of Angelica sinensis volatile oil was significantly higher than that of the model group.Decrease(P < 0.05);Conclusion:1.The abundance of Actinomyces in SHR was lower than that of Wistar rats,and that of Deferribacteres was higher than that of Wistar rats.There was a certain degree of disorder in the structure of intestinal flora in hypertensive rats.2.The volatile oil from Radix Angelicae Sinensis had no effect on the overall diversity and abundance of intestinal flora in spontaneously hypertensive rats.3.Volatile oil from Radix Angelicae Sinensis can significantly reduce the abundance of Proteus and Deferribacteres.4.The volatile oil of Radix Angelicae Sinensis has a certain intervention effect on the intestinal flora of SHR,possibly by regulating the abundance of Proteus and Deferribacteres。...
