The Protective Effect Of Longzhongxiaozhongzhitong Mixture On Secondary Spinal Cord Injury And Expression Of LncRNAMALAT1-ERK-p38MAPK-AQP-4 Axis

Objective Through the successful establishment of rat spinal cord injury model,the protective effect of Longzhongxiaozhongzhitong Mixture on spinal cord injury in rats and the effect of Longzhong Xiaozhongzhitong Mixture on the expression of lnc RNA MALAT1-ERK/p38 MA PK-AQP-4 axis were investigated.Methods1.A total of 175 adult male SD rats with clean grade were selected to establish the rat model of spinal cord injury by Allen’s method.2.After the establishment of SD rat model,the rats were randomly divided into control group(25 rats),sham injury group(25 rats),model group(25 rats),sodium escosaponin group(25rats),Longzhong Xiaozhongzhitong Mixture intervention group(75 rats),and Longzhong Xiaozhongzhitong Mixture intervention group(75 rats),which were divided into high dose group(25 rats),medium dose group(25 rats)and low dose group(25 rats).According to the BBB(Basso,Beattie & Bresnahan Locomotor Rating Scale,BBBSCAL)rating standard,scores were observed every 3 days and recorded for a total of 15 days.The water content of spinal cord was measured by wet and dry methods,and the damage of blood spinal cord barrier(BSCB)was quantitatively analyzed.After SSCI,the expressions of AQP-4,ERK,MALAT1 and p38 MAPK in spinal cord tissue were all increased,which was positively correlated with the degree of spinal cord injury.Longzhong Xiaozhongzhitong Mixture can relieve spinal cord edema by inhibiting the expression of AQP-4,ERK,MALAT1 and P38 MAPK in spinal cord tissue.Results1.Longzhong Xiaozhongzhitong Mixture can effectively treat the limb function of SD rats after secondary spinal cord injury.Compared with sodium aescinate,the effect of high dose of Longzhong detumescence on SSCI treatment was better than that of sodium aescinate and Longzhong detumescence zhitong mixture medium and low dose groups.2.Longzhong Xiaozhongzhitong Mixture can effectively relieve spinal cord edema and gradually restore the function of lower limbs in SD rats3.After SSCI,the expressions of AQP-4,ERK,MALAT1 and p38 MAPK in spinal cordtissue were all increased,which was positively correlated with the degree of spinal cord injury.Longzhong Xiaozhongzhitong Mixture can relieve spinal cord edema by inhibiting the expression of AQP-4,ERK,MALAT1 and P38 MAPK in spinal cord tissue.4.Longzhong detumescence and pain relieving mixture and sodium aescosaponin have therapeutic effect on SSCI,among which,both reduce the expression of AQP-4,MALAT1,ERK and p38 MAPK to reduce spinal cord edema and restore the function of lower limbs.Conclusion1.Longzhong Xiaozhongzhitong Mixture has the effect of preventing and treating SSCI.The main mechanism of its action is to restore the function of lower limbs and limbs of SD rats by reducing the compression of nerves caused by spinal edema.2.After SSCI,the blood-spinal cord barrier was destroyed,and the increased expressions of AQP-4,ERK,MALAT1,and p38 MAPK would aggravate the spinal cord injury,while the decreased expressions of AQP-4,ERK,MALAT1,and p38 MAPK would reduce spinal edema and restore limb function.3.Longzhong Xiaozhongzhitong Mixture can inhibit the expression changes of lnc RNA MALAT1-ERK/p38MAPK-AQP-4 axis.Through drug intervention,the expression of AQP-4,ERK,MALAT1 and p38 MAPK genes in the spinal cord tissue was reduced,the spinal cord edema and some neurological functions were alleviated,and the function of blood-spinal cord barrier was restored.4.After SSCI,sodium Aescinate and Longzhong Xiaozhongzhitong Mixture have curative effect on the prevention and treatment of SSCI.Through the comparison between the two drugs,Longzhong Xiaozhongzhitong Mixture has the effect of improving water and reducing swelling and reducing the expressions of AQP-4,MALAT1,ERK and p38 MAPK,which is better than that of sodium aescosaponin.
