| Objective:The aim of this study were to asses and compare the relative expression of miR-205-5p in serum exosome and serum among non-small cell lung cancer(NSCLC)patients,benign pulmonary conditions patients,and healthy volunteers respetivery.and to evaluate the clinical application value of serum exosome miR-205-5p as a tumor marker of non-small cell lung cancer.Methods:Serum samples were obtained between September 2019 and September 2020 from 22 NSCLC patients undergoing surgical treatment,20 patients diagnosed with a benign lung disease(lesion group),and 20 healthy volunteers(control group).Preoperative serum was provided in all NSCLC patients(preoperative group),but postoperative serum was provided in only 8 NSCLC patients(postoperative group).Quantitative RT-PCR was used to detect miR-205-5p expressions in serum and serum exosomes.Results:(1)The relative expression level of miR-205-5p from serum exosomes of NSCLC preoperative group was 5.744±2.519,which was significantly higher than that of benign lung disease group(1.330±0.220)and healthy volunteers(0.637±0.172)(both P <0.01).While there was no significant difference between the benign lung disease group and healthy volunteers group(P = 0.382).(2)Similarly,the relative expression level of miR-205-5p from serum of NSCLC preoperative group was2.527±0.874,which was significantly higher than that of benign lung disease group(0.880±0.132)and healthy volunteers(0.438±0.185)(both p<0.01,while there was no significant difference between the benign disease group and the healthy volunteers group(p=0.125).(3)Compared with the NSCLC preoperative group,the relative expression level of serum exosomal miR-205-5P and serum miR-205-5P in the NSCLC postoperative group were significantly decreased(both p<0.05).(4)relative expression level of miR-205-5p from serum exosomes were significantly higher than that from serum in the preoperative group,the postoperative group,the benign lung disease group and the healthy volunteers group(all P <0.01).Conclusion:(1)Mi R-205-5p derived from serum exosomes may be a novel tumor marker for NSCLC.(2)The relative expression level of serum exosomes miR-205-5p were higher than that of serum miR-205-5p,suggesting that serum exosomes miR-205-5p may be more suitable as tumor markers of NSCLC.(3)The expression level of serum exosomes miR-205-5p and serum miR-205-5p can reflect tumor burden. |
