Objective The purpose of this experiment was to investigate the effects of bone marrow mononuclear cell(BM-MNCs)transplantation on the expressions of mi R-21 and the inflammatory factor TNF-α in ulcerative colitis(UC)model mice,and to explore the possible mechanism of BM-MNCs transplantation in the treatment of UC.Methods Seventy-two qualified clean KM mice were randomly divided into three groups:transplantation group,PBS group and Control group,18 mice in each group,The ungrouped 18 are only used to extract BM-MNCs.Mice in transplantation group and PBS group were allowed to drink 2.5% dextran sodium sulfate(DSS)to prepare ulcerative colitie model.The mice in control group was routinely reared.After successful modeling,mice in the transplantation group were injected with BM-MNCs.The mice disease activity index(DAI)score and mucosal injury score of colon tissue were calculated.Expression of TNF-α in colon tissues was detected by Western blotting.Real-time PCR was used to detect the expression of mi R-21 in colon tissues。Results 1.The DAI scores of the mice in the transplantation group and the PBS group were higher than those in the control group,the difference was statistically significant(P<0.05),and the DAI scores of the mice in the transplantation group were significantly lower than those in the PBS group,and the difference was statistically significant(P <0.05).2.Control group had complete mucosa and orderly gland structure.In PBS group,a large number of neutrophils infiltrated colon tissue,crypt tissue was highly destroyed and glands were deformed.Inflammatory cell infiltration in colon tissue in transplantation group was relatively small,and inflammation was mild.The intestinal mucosal injury scores of transplantation group and PBS group were higher than those of Control group,and the difference was statistically significant(P< 0.05).The score of PBS group was higher than that of transplantation group,and the difference was statistically significant(P < 0.05).3.The levels of mi R-21 and TNF-α in the PBS group and the transplantation group were higher than those in the control group,with statistically significant differences(P <0.05).The levels of mi R-21 and TNF-α in the transplantation group were lower than those in the PBS group,with statistically significant differences(P <0.05).Conclusions 1.The ulcerative colitis model mice can be established by drinking DSS.2.BM-MNCs transplantation can down-regulate the expression of mi R-21 and TNF-α in the intestinal tissue of ulcerative colitis mice and improve intestinal inflammation.
