| BackgroundLung adenocarcinoma is a pathological tissue type with high incidence and poor prognosis in non-small cell lung cancer.It is of great significance to find new therapeutic targets for improving the prognosis of patients with lung adenocarcinoma.The fat mass and obesity associated gene(FTO),a member of the ALK B family,is a key enzyme for the removal of RNA m~6A methylation and plays a regulatory role in lipogenesis,spermatogenesis,development,cancer,stem cell regeneration and other diseases.However,the expression characteristics and clinical significance of FTO in lung adenocarcinoma tissues,as well as the role of FTO in tumor genesis and progression,are still not fully understood.ObjectiveTo investigate the protein expression of RNA demethylase FTO in lung adenocarcinoma tissues,and to analyze its correlation with clinicopathological features of lung adenocarcinoma and its relationship with prognosis of patients.MethodsThe expression level of FTO in lung adenocarcinoma tissues was predicted by TCGA database.Immunohistochemical method was used to detect the expression level of FTO protein in lung adenocarcinoma tissue microarray.The chi-square test was used to analyze the relationship between FTO protein expression and clinicopathological characteristics.Kaplan-Meier curve was used to analyze the relationship between FTO protein expression and prognosis.Results1.TCGA database found that the expression level of FTO m RNA in lung adenocarcinoma was significantly lower than that in adjacent tissues,and the overall survival rate of patients with high FTO expression was higher.2.Compared with paracancerous tissues,FTO protein was significantly higher expressed in lung adenocarcinoma tissues(20.4%and 45.4%,respectively),and the difference was statistically significant(P<0.05).The expression of FTO in lung adenocarcinoma tissues was positively correlated with age and pathological grade(P<0.05),was not related to the gender of the patient,TNM clinical stage,tumor size and lymph node metastasis(P>0.05).3.Kaplan-Meier curve analysis showed that the median survival time of patients with high FTO expression was 32 months,significantly lower than that of patients with low FTO expression(58 months)(P<0.05);Cox proportional risk model statistical analysis showed that low FTO expression(HR=1.573,P=0.041)was an independent factor for good prognosis in patients with lung adenocarcinoma;The Nomogram model was successfully constructed based on independent risk factors and showed good predictive power(C-index=0.728).ConclusionHigh expression of FTO protein levels in lung adenocarcinoma tissues is associated with poor prognosis of patients. |
