Objective:To explore the effect of sCD40L on proliferation and apoptosis of leukemic Nalm-6 cell line.Methods:(1)Detect the effect of different concentrations of sCD40L on the in vitro proliferation of Nalm-6 cells in acute lymphoblastic leukemia by CCK-8 method,and determine the optimal treatment concentration.(2)Apoptosis was detected by AnnnexinV-FITC/PI double staining.(3)Flow cytometry to detect the expression levels of related apoptosis factors Bax and Bcl-2 protein after sCD40L treatment of Nalm-6 cells.Results:(1)CCK-8 showed that different concentrations of sCD40L treated Nalm-6 cells for 24h,48h,and 72h,all had different degrees of inhibition,which was compared with the dexamethasone sodium phosphate group(positive control group)and Nalm-6 cell group(negative control group)In comparison,there are significant differences(P<0.05);the cell proliferation inhibition rate gradually increases with time,and the cell inhibition rate is most obvious at 72h;the cell proliferation inhibition rate gradually increases with the increase in concentration,at 5.00μg/ml The proliferation inhibition rate of sCD40L was the most obvious at 72h;however,the cell proliferation inhibition rate of sCD40L at 4.00μg/ml and 5.00μg/ml was not statistically significant(P>0.05),so the concentration of 4.00μg/ml was chosen as the follow-up experiment.(2)Double staining with AnnexinV-FITC/PI showed that Nalm-6 cells were treated with sCD40L at 24h,48h,and 72h.At 48h,sCD40L had the most obvious effect on the apoptosis rate of Nalm-6 cells;it was compared with Nalm-6 cell group and dexamethasone phosphate.Compared with sodium group,there were significant differences(P<0.05).(3)Flow cytometry showed that the expression of Bax protein was significantly different at 24h,48h,72h after 24h,48h,and 72h after sCD40L treatment of Nalm-6 cells(P<0.05),and the up-regulation was most obvious at 48h.The expression of Bcl-2 protein was significantly different at 24h,48h,and 72h(P<0.05),and the down-regulation was most obvious at 48h.Conclusion:(1)sCD40L can inhibit the proliferation of Nalm-6 cells and induce their apoptosis,which is related to the concentration and time;(2)The apoptosis of Nalm-6 cells induced by sCD40L may be related to the mitochondrial signal pathway mediated by Bax/Bcl-2.
