| Objective: To analyze the effects of LSD1 on Wnt/β-catenin pathway activity,proliferation and invasion of colon cancer cells in vitro.Method:1.The HCT116 and HT29 cells that LSD1 gene were overexpressed or silenced by cell transfection technology.The expression of β-catenin,E-cadherin,MMP9 and Slug were detected by Western blot,the Wnt/β-catenin pathway activity was detected by TOP-Flash luciferase reporter gene experiment,cell proliferation and invasiveness ability were detected by MTT and Transwell experiment,respectively,To analyze the effects of LSD1 on Wnt/β-catenin pathway activity,proliferation and invasion of colon cancer cells.2.Wnt/β-Catenin pathway agonists or inhibitors were used to interfere with LSD1 gene overexpressed or silenced HCT116 and HT29 cells,the expression of β-catenin,E-cadherin,MMP9 and Slug were detected by Western blot,the Wnt/β-catenin pathway activity was detected by TOP-Flash luciferase reporter gene experiment,cell proliferation and invasiveness ability were detected by MTT and Transwell experiment,respectively.To analyze the role of Wnt/β-catenin pathway in the proliferation and invasive ability of colon cancer cells affected by LSD1.Results:1.Compared with the blank control group and the negative control group,the expression of β-catenin,MMP9 and Slug in the HCT116 and HT29 cells of the LSD1 silenced group was significantly down-regulated,the expression of E-cadherin was significantly up-regulated,Wnt/β-Catenin pathway activity,proliferation and invasiveness of cells were significantly down-regulated.the difference was statistically significant(P<0.05),the expression of β-catenin,MMP9 and Slug in HCT116 and HT29 cells of the LSD1 overexpressed group was significantly up-regulated,the expression of E-cadherin was significantly down-regulated,Wnt/β-Catenin pathway activity,proliferation and invasiveness of cells were significantly up-regulated;the difference also was statistically significant(P<0.05).2.Rescue experiments showed that compared with the group of LSD1 gene silenced,The inhibitory effect of LSD1 gene silenced on Wnt/β-catenin pathway activity,proliferation and invasion ability of cells could be partially reversed after intervention with Wnt/β-Catenin pathway agonist.the difference was statistically significant(P<0.05).Compared with the group of LSD1 gene overexpressed,after adding the Wnt/β-Catenin pathway inhibitor,the stimulating effect of LSD1 gene overexpression on Wnt/β-Catenin pathway activity,proliferation ability and invasiveness of cells can partially reverse,the difference also was statistically significant(P<0.05).Conclusion:1.The Wnt/β-catenin pathway activity,proliferation and invasion ability of HCT116 and HT29 cells can be inhibited by silenced of LSD1 gene.at the same time,it can also be up-regulated by overexpressed of LSD1 gene.2.The functional recovery experiment confirmed that the effect of LSD1 on proliferation and invasion of HCT116 and HT29 cells could be mediated by Wnt/β-catenin pathway. |
