Objective:By observing the morphology of rat nasal mucosa,analyzing rat serum and nasal mucosal Th1,Th2,Th17 cell immune indicators,and exploring the effect of Qifenggubiao granule on Th1,Th2,Th17 cell immune function of allergic rhinitis(AR)rats Influence,further clarify the mechanism of action of Qifenggubiao granule in the treatment of AR.It provides experimental basis for the study of Qifenggubiao granule in the treatment of AR,and provides other reliable and scientific treatment ideas for the clinical treatment of AR.Methods:60 SPF male SD rats were randomly divided into 6groups,each with 10 rats,divided into blank group,model group,positive control group,Qifenggubiao granule high-dose group,Qifenggubiao granule medium-dose group,Qifenggubiao granule low-dose group.Ovalbumin and aluminum hydroxide were used for intraperitoneal injection for 14 days,and then ovalbumin solution was used for nasal enhancement spurs for 7 days to establish an AR model.After the sensitization,the superimposed quantitative scoring method was used to score the nasal symptoms of the rats.If the score is more than 5 points,it indicates that the model is successful.After the model was successfully established,the drug was continuously administered for 14 days.After the gavage,serum and nasal mucosa of rats were collected.The hematoxylin-eosin staining method(HE)was used to observe and compare the pathological changes of the nasal mucosal sections of the rats in each group;the enzyme-linked immunoassay(ELISA)was used to determine the levels of IFN-γ,IL-4,and IL-17 in the serum of each group of rats;reverse transcription polymerase chain reaction(RT-PCR)was used to detect T-bet and GATA-3 and the relative expression level of RORγt m RNA.Results:Compared with the blank group,rats in the model group showed typical symptoms of allergic rhinitis,such as scratching the nose,scratching the face,rhinorrhea,and sneezing.The score of nasal symptoms was significantly increased,and the difference was statistically significant(P<0.01);Pathological examination found that compared with the blank group,the nasal mucosa of rats in the model group was significantly swollen,accompanied by a large number of eosinophil infiltration;compared with the model group,the nasal mucosa of the rats in the Qifenggubiao granule high and medium dose group was swollen And the infiltration of eosinophils was significantly reduced,and there was no significant difference between the high-dose group and the positive control group.The content of each cytokine in the rat serum was measured by ELISA and it was found that compared with the blank group,the IFN-γ in the serum of the model group was The content of was significantly reduced(P<0.01),and the content of IL-4 and IL-17 were significantly increased(P<0.01);compared with the model group,the content of IFN-γ was significantly increased in the Qifenggubiao granule high and middle dose groups(P<0.01),the content of IL-4 and IL-17 was significantly reduced(P<0.01).With the increase of the dose,the effect on the increase of IFN-γcontent and the decrease of the content of IL-4 and IL-17 increased The effect of the Qifenggubiao high-dose group is equivalent to that of the positive control group;the relative expression level of genes in the nasal mucosa of rats was detected by PT-PCR and it was found that compared with the blank group,the relative expression level of T-bet m RNA in the model group Down-regulation(P<0.01),relative expression levels of GATA-3m RNA and RORγt m RNA up-regulation(P<0.01);compared with the model group,the relative expression levels of T-bet m RNA in each dose group of Qifenggubiao granules were up-regulated,and GATA-3 The relative expression levels of m RNA and RORγt m RNA were down-regulated,and as the dose increased,the relative expression levels of T-bet m RNA were up-regulated and the relative expression levels of GATA-3 m RNA and RORγt m RNA were down-regulated.Conclusion:Qifenggubiao granules can significantly improve the typical symptoms of allergic rhinitis in rats,such as nasal scratching,sneezing,runny nose and listless spirit,and can effectively promote the repair of nasal mucosa,and can significantly reduce the pathological damage in AR rats.Qifenggubiao granules can effectively treat allergic rhinitis,and its mechanism is realized by regulating the differentiation of Th1,Th2 and Th17 cells.
