| The"Chinese Pharmacopoeia"stipulates that the medicinal material of Hyoscyamus is the dried mature seeds of Solanaceae Hyoscyamus niger L..It has antispasmodic,pain relief,anti-asthmatic and soothing effects.This article refers to the research methods of traditional botanical pesticides,Study on the active ingredients and antibacterial activity of Hyoscyamus seeds.The main results are shown as follows:1.Isolation and structure elucidation of active ingredients:Application of separation methods such as silica gel column chromatography,Sephadex LH-20,reversed-phase chromatography,10 compounds were separated from the n-butanol layer of the 95%ethanol extract of Hyoscyamus serrata seeds.Through various spectroscopic techniques,The structures of 9 compounds were identified through various spectroscopic techniques.including4 alkaloids:Cronamide(6A001D),N-trans-ferulamide(6A007-37),Cannabiamide D(6A027B)and Hyoscyamine(6A038B),3 aromatic derivatives:dibutyl phthalate(6A014F),vanillic acid(6A0XCS)and vanillin(6A0XCQ),1 flavonoid glycoside:quercetin-3-O-β-D-rutin(6A0LD),1 glycerin(6A038A).Among them,the structure of 6A011C has not been characterized yet.2.Fungal inhibitory activity assay:Evaluated the antibacterial activity of 5 monomer compounds 6A001D,6A014F,6A0LD,6A0XCS,6A0XCQ.The 11 types of fungi are Alternaria mali Rob.,Macrophoma kawatsukai Hara,Phytophthora capsica Leonian,Sclerotinia sclerotiorum(Lib.)de Bary,Coniothyrium diplodiella(Speq.)Sacc,Alternaria solai(Ell.et Mart.)Jones et Grout.,Botrytis cinerea Pers.Fr.,Valsa mali Miyabe et Yamada.,Rhizoctonia solani Kühn,Alternaria alternata(Fr.)Keissler,Alternaria alternata(Fr.)Keissler and Colletotrichum capsici(Syd.)Butl.the result shows:6A014F has good antibacterial activity against on Sclerotinia sclerotiorum(Lib.)de Bary,6A0XCQ shows good antibacterial activity on Valsa mali Miyabe et Yamada.Its EC50 are 117.509(?)g/m L and 102.658(?)g/m L,respectively.3.Bacterial inhibitory activity assay:Evaluated the antibacterial activity of 5 monomer compounds 6A001D,6A014F,6A0LD,6A0XCS,6A0XCQ.The test bacteria are Pseudomonas syringae pv.actinidiae,Xanthomonas campestris pv.citri(Hasse)Dye,Erwinia carotovora,Ralstonia solanacearum and Ralstonia solanacearum(E.F.Smith)Yabuuhi et.the result shows:The five monomer compounds have no significant antibacterial activity against the five bacteria.4.Tobacco mosaic virus(TMV)inhibition assay:Determining the inactivation and inducing effects of nine kinds of monomer compounds on TMV,The 9 monomer compounds are 6A001D,6A07-37,6A011C,6A014F,6A0LD,6A0XCS,6A0XCQ,6A038B and 6A027B The result shows:the monomeric compound 6A027B has 100%inducing effect on TMV,6A0XCS,6A0XCQ have obvious inactivation effect on TMV.At concentrations of 500(?)g/m L,250(?)g/m L and 125(?)g/m L,the inhibition rates were 79.71%,62.50%,47.30%,54.37%,43.21%,and 34.78%.6A011C has obvious inducing effect on TMV,and the inhibition rates were 69.14%,58.00%,and 20.57%at 500(?)g/m L,250,anvd 125(?)g/m L,respectively.The above research results clarify the agricultural antibacterial and antiviral active substance basis of Hyoscyamus serrata seeds.It laid a theoretical foundation for its further development as plant-derived fungicides and anti-phytoviral agents. |
