| ObjectioveTo select long non-codingRNA(lncRNA)related to survival of glioma patients and predict its possible regulatory target genes as well as related signal pathways,and explore its prognostic value through bioinformatics analysis.MethodMining the differentially expressed lncRNA,in glioma with the Gene Expression Omnibus(GEO)and analyzing the survival of these differentially expressed lncRNA via GEPIA,the database based on The Cancer Genome Atlas(TCGA),and then through the joint analysis of mi Rcode,mi RDB,mi RTar Base,and Target Scan to discover the mRNA and signal pathways related to differentially expressed lncRNA.Finally,investigate its function by gene enrichment analysis.In order to further understood its role in Chinese patients,survival analysis,expression analysis,Cox proportional hazard analysis and GSEA analysis were performed based on CGGA database.Result23 lncRNAs were discovered differentially expressed in gliomas by GEO,of which 10 were down-regulated 13 were up-regulated.Online differential expression analysis and survival analysis performed on GEPIA found that the down-regulated expression of two lncRNAs MIR7-3HG and RFPL1 S in TCGA was positively related to overall survival and disease-free survival of glioma patients.Further target gene prediction showed that lncRNA RFPL1 S was potentially related to 23 miRNA targets and 952 mRNA targets,while gene enrichment analysis of these mRNA targets showed that they were enriched in many confirmed tumor related pathways,such as MAPK signal pathway,PI3K-Akt signal pathway,TGF-β signal pathway,Erb B signal pathway and so on.Enrichment analysis based on TCGA showed that lncRNA RFPL1 S can serve as an independent prognostic factor of low-grade gliomas and participate in NF-κ B signal pathway and other signal pathways related to glioma invasion and proliferation.CGGA survival analysis had also proved lncRNA RFPL1 S to be positively related to survival of glioma patients;COX proportional hazard analysis suggested that lncRNA RFPL1 S could serve as an independent prognostic factor of glioma;while differential expression analysis suggested that lncRNA RFPL1 S was differentially expressed in gliomas of different ages,pathological types and pathological grades.GSEA enrichment analysis showed that lncRNA RFPL1 S coexpressed genes were enriched in GAP Junction signal pathway and Gn Rh signal pathway,suggesting that lncRNA RFPL1 S may be involved in the occurrence and invasion of glioma.ConclusionThrough joint bioinformatic analysis based on multiple databases,a lncRNA RFPL1 S,was discovered to be positively related to survival of glioma patients.The mRNA targets or co-expressed genes of lncRNA RFPL1 S are enriched in multiple tumor-related signal pathways.COX proportional hazard analysis showed that lncRNA RFPL1 S could serve as an independent prognostic factor for gliomas.Our study provides a potential biomarker to further understand the mechanism,diagnosis,treatment and prognosis of gliomas. |
