| Objective: To study the effects of gastrodin injection on bone mineral density,bone biomechanics and related bone metabolism indexes in osteoporotic rats after ovariectomy.Methods: Forty healthy female unborn Sprague Dawley(SD)rats were selected as the research objects.According to the random number sequence method,they were random Ly divided into 4 groups,namely the sham operation group,the model group,the estradiol group,and the gastrodin group.The model group,the estradiol group,and the gastrodin group were surgically removed from both ovaries,while the sham operation control group was removed from both sides of the ovarian adipose tissue.The gastrodin group was given intraperitoneal injection of gastrodin injection,the estradiol group was given estradiol benzoate,and the sham operation group and model group were given equal volume of normal saline,once a day.After the intervention was completed12 weeks,the body weight and wet weight of the right femur of each group of rats were observed and recorded,and related tests such as bone metabolism index,femoral bone density,bone biomechanics and bone tissue morphology were performed.Results:1.Rat body weight changes: There was no significant difference in the weight of rats in each group before operation(P > 0.05).After 12 weeks of intervention,the weight of the model group was significantly higher than that of the sham operation group,estradiol group,and gastrodin group,with statistical differences(P < 0.05)2.Rat femur wet weight: Compared with the sham operation group,the wet weight of the right femur of the rats in the other groups was reduced,with statistical difference(P < 0.05);Compared with the model group,the wet weight of the right femur of the rats in the estradiol group and the gastrodin group increased,and there was a statistical difference(P < 0.05).3.Bone metabolism indexes: Compared with the sham operation group,the rats in the model group have higher serum osteocalcin,lower blood calcium and blood phosphorus levels,with statistical differences(P < 0.05);Compared with the model group,the rats in the estradiol group and gastrodin group were lower in serum osteocalcin and higher in blood calcium and phosphorus,with statistical differences(P < 0.05).4.Femoral bone mineral density: Compared with the sham operation group,the femoral bone mineral density of rats in the model group was reduced,with statistical difference(P < 0.05);Compared with the model group,the femoral bone mineral density of rats in the estradiol group and gastrodin group increased,and there was a statistical difference(P < 0.05).5.Bone biomechanics: Compared with the sham operation group,the maximum load and bending strength of the femur in the model group were reduced,and there was a statistical difference(P < 0.05);Compared with the model group,the maximum load and bending strength of the femur of rats in the estradiol group and gastrodin group were increased,and there was a statistical difference(P < 0.05).6.Femoral bone tissue morphology: Compared with the sham operation group,the trabecular bone of the model group was significantly reduced,thinner,sparsely arranged,and partially broken;Compared with the model group,the number of bone trabeculae in the estradiol group and the gastrodin group increased,the arrangement was more orderly,and the continuity could be improved.Conclusions:1.Gastrodin can increase the bone density of osteoporotic rats after ovariectomy,improve the biomechanical properties of bone,and affect related bone metabolism indicators.2.This experiment suggests that gastrodin has a certain preventive effect on osteoporosis caused by ovariectomized rats. |
