Correlation Between Serum ANGPTL8 Levels And Atherosclerosis In Initial Type 2 Diabetes

Objective:The purpose of this study was to investigate the difference between newly diagnosed type 2 diabetes patients(T2DM)and healthy people in serum angiopoietin-like protein 8(ANGPTL8)level,the difference of ANGPTL8 between different carotid intima-media thickness(CIMT)groups and healthy people.And further test whether ANGPTL8 has atherosclerotic effect on T2 DM individuals.Methods:A total of 161 subjects were included in this study,including 83 initial T2 DM patients and 78 healthy controls.Height,weight,blood glucose,insulin,C-peptide,Glycosylated hemoglobin(Hb A1c),blood lipid and serum ANGPTL8 levels of all subjects were measured,and carotid intima-media thickness(CIMT)was measured under ultrasound.the relationship of ANGPTL8 with T2 DM and CIMT thickness,the risk factors affecting CIMT thickness in T2 DM population was analyzed.Results: 1.Compared with healthy control group,serum ANGPTL8 level in T2 DM group was significantly higher.In healthy control group,T2 DM with normal CIMT group and T2 DM with thickened CIMT group,serum ANGPTL8 showed a gradually increasing trend;2.Correlation analysis showed that serum ANGPTL8 level of T2 DM patients was positively correlated with age(r=0.235,p=0.033),FCP(r=0.25,P=0.023),Ty G(r=0.456,p<0.01),CIMT(r=0.393,p<0.01)and TG(r = 0.538,p<0.01).negatively correlated with HDL(r=-0.198,P=0.048).TG is an independent risk factor for the increase of serum ANGPTL8(OR=3.175,p =0.027);3.In T2 DM patients,CIMT was positively correlated with ANGPTL8(r=0.393,p<0.01)and TG(r = 0.24,p < 0.05).After adjusting for age and course of T2 DM,ANGPTL8 level was still independently correlated with CIMT(r= 0.336,P = 0.002),while no significant correlation was found with other indicators;4.Taking CIMT thickness as a dependent variable,after adjusting for age,course of T2 DM,FCP,TG,HDL and other factors,binary Logistic regression analysis showed that serum ANGPTL8 was a risk factor for CIMT thickening.Conclusion:1.Serum ANGPTL8 level was significantly increased in T2 DM patients;2.In healthy control group,T2 DM group with normal CIMT group and T2 DM with thickened CIMT group,serum ANGPTL8 level showed a gradually increasing trend;3.Serum ANGPTL8 is involved in the occurrence and development of AS in T2 DM patients,and is a risk factor for cardiovascular diseases in T2 DM patients;4.ANGPTL8 is closely related to TG metabolism,and TG is supposed to play a mediating role in the correlation between ANGPTL8 and CIMT.
