Objective:Study the expression of immune checkpoint molecules T cell immunoglobulin and ITIM domain(TIGIT)in gliomas,and preliminarily explore their relationship with clinicopathological characteristics and prognosis.Methods:According to the exclusion criteria,the tumor tissues of 54 patients with glioma who underwent surgical treatment at the First Hospital of Shanxi Medical University from October 2018 to October 2020 and the pathological diagnosis was clear and 18 brain tissues of non-tumor patients who underwent craniotomy were obtained.TIGIT staining was performed by immunohistochemical staining method to observe its expression in gliomas,and then using immunofluorescence staining methods to determine its expression in immune cells,and analyze the relationship between the expression of TIGIT and prognosis,clinicopathological characteristics with the patient’s age,gender,and tumor size,Pathological grade,IDH,KPS score.Results:1.TIGIT lowly expressed in non-tumor brain tissues positively rated only 11.1%.while reach 83.3% in glioma tissue.The Brown strongly stained part is mainly located on the T cell membrane.Compared with low-grade gliomas,TIGIT expression is higher in high-grade gliomas.In the semi-quantitative analysis of the TIGIT expression score of gliomas,the expression level of TIGIT in high-grade and low-grade gliomas increased significantly(P<0.01).2.The expression of TIGIT protein in the glioma group was significantly related to the pathological grade of the patient(P=0.025),and the overexpression of TIGIT was related to the high pathological grade of glioma.The expression of TIGIT is correlated with IDH mutation(P=0.001).IDH mutants tend to have lower TIGIT expression in gliomas.However,there was no statistical significance in comparison with the patient’s gender,age,tumor size,and KPS score(P>0.05).3.Although TIGIT is related to the pathological grade of glioma,its expression in glioma is not statistically significant compared with the patient’s prognosis.However,it is worth noting that there is a statistically significant difference in OS and DFS between TIGIT and glioma patients in GBM(P<0.05).Conclusion:The expression of TIGIT in glioma tissue is higher than that in non-tumor tissues.As the grade of glioma increases,the expression of TIGIT gradually increases,but it is low in non-tumor tissues.The expression of TIGIT in glioma tissue is related to the patient’s pathological grade and IDH mutation,and has nothing to do with the patient’s age,gender,tumor size,and KPS score.The expression of TIGIT is related to the prognosis of GBM,and the high expression of TIGIT indicates poor prognosis of GBM.
