| Objective:In this study,we used bioinformatics methods to comprehensively and deeply study the expression and prognostic significance of autophagy-related lncRNAs in hepatocellular carcinoma,screened out valuable lncRNAs,constructed prognostic risk score,and combined with clinical characteristics to construct prognostic nomogram to predict the survival rate of patients,and preliminarily explored and predicted the role of autophagy-related lncRNAs in hepatocellular carcinoma and autophagy pathway through ce RNA regulatory network.Methods:Using bioinformatics methods,we obtained the RNA expression data,survival information and clinical characteristics of HCC patients from TCGA database,and downloaded the list of autophagy-related genes from HADb website.The differential expression of autophagy-related lncRNA was identified by differential expression analysis and co-expression analysis.Cox regression survival analysis and LASSO analysis were used to screen the autophagy-related lncRNA that affect the prognosis,then the prognostic risk score system was constructed.Kaplan-Meier survival analysis and ROC curves were used to evaluate the predictive ability,and the correlation between risk score and different clinical features was studied.The prognostic factors of HCC patients were included in the construction of nomogram to predict the 1-and 3-year survival rate.The accuracy and reliability of nomogram prediction results were evaluated by consistency index,correction curve and ROC curves.Based on autophagy-relatedlncRNAs,the targeted relationship among lncRNA-mi RNA-m RNA was predicted in Starbase database,and the ce RNA regulatory network was constructed.Results:1.A group of autophagy-related lncRNAs(DEARlncRNAs)were identified by differential expression analysis and co-expression analysis.A prognostic risk score system consisting of 7 lncRNAs was constructed by screening the prognostic value of DEARlncRNAs.The risk score is an independent risk factor for the prognosis of HCC patients.The risk score is significantly correlated with pathological grade and TNM stage,and with the increasing of grade and stage,and the higher the score is,the worse the prognosis is;2.Using risk score and TNM stage to construct prognostic nomogram can easily and accurately predict the 1-and 3-year survival probability of patients with HCC;3.Based on the risk score of 7 autophagic lncRNAs with important prognostic value,a ce RNA regulatory network composed of 4 DEARlncRNAs,8 DEmi RNAs and 15 DEARGs was constructed.Conclusion:1.Based on autophagy-related lncRNAs,a nomogram was constructed to predict the prognosis of HCC,which can accurately and reliably predict the 1-year and 3-year survival rate of patients;2.Based on autophagy-related lncRNAs,a regulatory network of ce RNA was constructed in HCC composed of 4 lncRNAs,8 mi RNAs and 15 m RNAs,which has important research value. |
