Preliminary Study On The Relationship Between Bem46 And Cell Wall Related Genes In Aspergillus Fumigatus

Objective: The expression of cell wall related genes KTR1、KTR4、KTR7、GPI7、MSDS gene between Aspergillus fumigatus standard strains and Bem46-knockout strains was testing by RT-PCR,the relationship between KTR1、KTR4、KTR7、GPI7、MSDS gene and Bem46 affect polar growth was analyzed.Methods: 1.Activate the Aspergillus fumigatus standard strains,Ku80 and ΔBem46 strains in the laboratory;2.Make relevant culture medium;3.Search the sequences of Bem46、KTR1、KTR4、KTR7、GPI7、MSDS and internal reference genes β-tubulin in NCBI,design and synthesize primers;4.Total RNA was extracted and reverse transcribed into c DNA,and the RT-PCR was used to detect the expression of each gene in standard strain and knockout strain.Results: By two independent samples t-test,the expression levels of KTR1 and MSDS had no statistical significance(P>0.05);the expression levels of KTR4、KTR7、GPI7 had statistical significant(P<0.05),which are 0.69 times、0.31 times and 4.45 times respectively in ΔBem46 and Ku80.Conclusion:In ΔBem46,the expression of KTR4、KTR7 were decreased,while the expression of GPI7 was increased.The expression of KTR1 and MSDS had no apparent different compared with Ku80.We speculated that Bem46 gene may participate in the polar growth process in A.fumigatus by participating in polymerization process of galactomannan and influencing the fixation process of related proteins in the cell wall.
