| Object: To explore the effect of circular RNA(circ RNA)hsa_circ_0000326 on the biological function in glioma and its clinical significance.Methods: Firstly,in order to find a circ RNA,we analyzed the differential circ RNAs expression of glioma through a circ RNA chip(GSE146463)in the GEO database.Real-Time Quantitative Reverse Transcription Polymerase Chain Reaction(q RT-PCR)was used to detect the expression levels of hsa_circ_0000326 in glioma samples of 38 patients and normal brain tissues of 16 patients with traumatic brain injury.Meanwhile,we collected the clinical data onto patients,analyzing by Fisher’s exact test,Kaplan-Meier plot and COX regression model,to determine the relationship between hsa_circ_0000326 levels and prognosis of glioma patients,and explore its clinical significance.Secondly,hsa_circ_0000326 knockdown model was constructed using lentivirus and transfect in U251 and U87 cells.To investigate the impact of hsa_circ_0000326 on the biological function of glioma,we utilized many techniques,such as Cell Counting Kit-8(CCK-8),cell clone formation,cell scratch healing,Transwell chamber invasion and migration,flow cytometry,acridine orange(AO)staining and Western blot(WB).Finally,we used online databases and Cytoscape software to predict and construct a circ RNA-mi RNA-m RNA regulatory network.Results: qRT-PCR results showed that the expression level of hsa_circ_0000326was conspicuously increased in glioma tissues compared with normal brain tissues,and it was related to tumor grade.Kaplan-Meier curve showed that the overall survival of patients in hsa_circ_0000326 high expression group is relatively short.The COX regression suggested that the overexpression of hsa_circ_0000326(HR=5.462,95% CI(1.077-27.691),P=0.040)was a risk factor affecting survival in glioma patients.Cell experiments indicated that hsa_circ_0000326 knockdown could inhibit the proliferation,invasion,migration and autophagy of U251 and U87 cells,as well as promote cell apoptosis.Conclusions: Hsa_circ_0000326 was up-regulated in glioma tissues.It could promote the proliferation,invasion,migration and autophagy of glioma cells,as well as inhibit cell apoptosis Up-regulation of hsa_circ_0000326 is positively relevant to poor prognosis of glioma patients.And it might serve as a biomarker and target for clinical treatment of glioma.Hsa_circ_0000326 may function as competing endogenous RNAs to regulate the progression of glioma. |
