Objective:To discuss the protein and mRNA expression of Cx43(Connexin 43),mTOR(Mammalian target of rapamycin)and p70S6K(Ribosomal Protein S6 Kinases 70-kDa)in gastric cancer and normal gastric mucosal tissue and explore the clinical significance and the interactions of them.Materials and methods:From February 2019 to September 2019,60 cases of gastric adenocarcinoma specimens resected by radical surgery in the Affiliated Hospital of Chengde Medical College were collected.Two soy-sized masses and two normal gastric mucosa tissues(considered normal gastric mucosa tissues at least 5cm away from the cancer focus)were taken from the fresh specimen,part of which was made into wax blocks,and the otherpart was stored in the refrigerator at-80℃.Using SP Immunohistochemistry and Western Blotting technology to test the protein expression of Cx43,mTOR and p70S6K,and qRT-PCR technique to detect the mRNA expression of Cx43,mTOR and p70S6K in gastric cancer tissues and normal mucosal gastric tissues.The obtained data were analyzed by SPSS 25.0 statistical software,and correlation analysis was performed by Pearson method.P<0.05 was considered statistically significant.Results:1 The protein and mRNA expression of Cx43,mTOR and p70S6K in normal gastric mucosal tissues and gastric cancer mucosal tissuesIn the mucosal tissues of gastric cancer,the protein and mRNA expression of Cx43 were obviously lower than normal(all P<0.05);on the contrary,the protein and mRNA expression of mTOR and p70S6K were obviously higher than normal(all P<0.05).2 The relationship between clinic pathological indicators and the protein and mRNA expression of Cx43,mTOR and p70S6K in gastric cancer mucosal tissuesIn gastric cancer mucosal tissues,the protein and mRNA expression of Cx43,mTOR and p70S6K have no connection with sex and age(all P>0.05).The protein and mRNA expression of Cx43 protein of poorly differentiated,Ⅲ and Ⅳ stage of TNM,infiltrate beyond the muscular layer and lymphatic metastasis in gastric mucosal cancer were lower than well-mederately differentiated,Ⅰ and Ⅱ stage of TNM,the infiltration does not exceed the muscular layer and no lymphatic metastasis in gastric cancer(all P<0.05).The protein and mRNA expression of mTOR and p70S6K of poorly differentiated,Ⅲ and Ⅳ stage of TNM,infiltrate beyond the muscular layer and lymphatic metastasis in gastric mucosal cancer were higher than well-mederately differentiated,Ⅰ and Ⅱ stage of TNM,the infiltration does not exceed the muscular layer and no lymphatic metastasis in gastric mucosal cancer(all P<0.05).3 Correlation analysis of Cx43,mTOR and p70S6KThe relationship between the protein expression levels of the three detection indexes in immunohistochemistry,Cx43 was negatively correlated with mTOR(r=-0.362);Cx43 was negatively correlated with p70S6K(r=-0.318);mTOR was postively correlated with p70S6K(r=0.362).(all P<0.05)The relationship between the protein expression levels of the three detection indexes in western blotting,Cx43 was negatively correlated with mTOR(r=-0.531);Cx43 was negatively correlated with p70S6K(r=-0.443);mTOR protein was postively correlated with p70S6K(r=0.780).(all P<0.05)The relationship between the mRNA expression levels of the three detection indexes in qRT-PCR,Cx43 was negatively correlated with mTOR(r=-0.528);Cx43 was negatively correlated with p70S6K(r=-0.667);mTOR was postively correlated with p70S6K(r=0.458).(allP<0.05)Conclusion:1.In gastric cancer mucosal tissues,the expression level of Cx43 was lower than normal,on the contrary,the expression levels of mTOR and p70S6K were significantly higher than normal,mTOR and p70S6K play a synergistic role in the occurrence of cell biological processes.2.In the process of the occurrence of gastric cancer development,the expression of Cx43 and mTOR/p70S6K are negatively correlated,may be the Cx43 protein expression in gastric cancer tissue are reduced or absent,the Cx43 mediated by the GJIC function decline,directly or indirectly lead to excessive activation of mTOR/p70S6K signal pathways,resulting in the occurrence of gastric cancer development,thus raising Cx43 inhibits the development of cancer of the stomach,for potential drug targets and targeted therapy of gastric cancer pathogenesis provides a new train of thought.
