| Objective:For the past few years,more and more attention has been paid to skin tumors.Its incidence rate is increasing year by year,which not only affects personal physical and mental health,but also seriously increases the social burden.Therefore,it is of great significance to study the pathogenesis and the key factors of malignant transformation of skin tumors.In this study,the expression of Ets2,ROCK2,eIF4 E and P-4EBP1 in SK,BD and cSCC was studied,and the significance and correlation of each index in skin tumor were discussed,so as to provide theoretical reference for clinical diagnosis and treatment.Methods:1.Tissue wax blocks were provided by the Department of Dermatology of the Affiliated Hospital of Chengde Medical College and Cangzhou Central Hospital,which were confirmed by two senior pathologists.There were 50 cases of SK tissue,16 cases of BD tissue,28 cases of cSCC tissue and 10 cases of normal skin tissue.Immunohistochemical stain was used to detect the expression of Ets2,ROCK2,eIF4 E and P-4EBP1 in the above groups.2.The experimental data were processed by SPSS 25.0 software.Pearson test,continuity correction test or Fisher’s exact probability method were used to compare the positive rates.Spearman rank correlation analysis was used to compare the correlation between Ets2,ROCK2,eIF4 E and P-4EBP1,and the difference was statistically significant when P < 0.05.Results:1.The positive expression rates of Ets2 in cSCC group,BD group,SK group and normal tissue were 92.86%(26/28),75.00%(12/16),46.00%(23/50)and 0(0/10)respectively.After pairwise comparisoned between groups,cSCC and BD groups were higher than SK group and normal group(P < 0.05),However,there was no statistical significance between cSCC and BD group(P >0.05).The SK group was higher than normal group(P < 0.05).2.The positive expression rates of ROCK2 in cSCC group,BD group,SK group and normal tissue were 96.43%(27/28),68.75%(11/16),40.00%(20/50)and 0(0/10)respectively.After pairwise comparisoned between groups,cSCC group was higher than BD group,SK group and normal group.The BD group was higher than SK group and normal group.The SK group was higher than normal group.(all P <0.05).3.The positive expression rates of eIF4 E in cSCC group,BD group,SK group and normal tissue were 100%(28/28),75.00%(12/16),42.00%(21/50)and 0(0/10)respectively.After pairwise comparisoned between groups,cSCC group was higher than BD group,SK group and normal group.The BD group was higher than SK group and normal group.The SK group was higher than normal group.(all P <0.05).4.The positive expression rates of P-4EBP1 in cSCC group,BD group,SK group and normal tissue were 71.43%(20/28),68.75%(11/16),38.00%(19/50)and 0(0/10)respectively.After pairwise comparisoned between groups,cSCC and BD groups were higher than SK group and normal group(P< 0.05),However,there was no statistical significance between cSCC and BD group(P >0.05).The SK group was higher than normal group(P < 0.05).5.There was a significant positive correlation between eIF4 E and P-4EBP1 positive expression rates in SK and BD tissues(all P <0.05),and there was a significant positive correlation between Ets2 and P-4EBP1 positive expression rates in cSCC tissues(P <0.05).Conclusion:1.Ets2 may plays an important role in the occurrence and development of cSCC,BD,SK,and may have reference significance for the malignant transformation of the above tumors,but it has no predictive significance for the progression of BD to cSCC.2.ROCK2 may plays a very important role in the occurrence and progression of cSCC,BD,SK,and may promote keratinocytes malignant transformation,promote the invasion and metastasis of skin squamous cell carcinoma in situ,and provide a theoretical reference for the treatment and prognosis of epithelial proliferative tumors.3.eIF4 E plays a key role in the occurrence,malignant transformation and even invasion process of cSCC,BD,SK.It is speculated that eIF4 E can not only promote the deterioration,invasion and metastasis of benign keratinocytes,but also predict the malignant degree of epithelial-derived tumors.4.P-4EBP1 may promote the proliferation and malignant transformation of benign epidermal tumors,but it cannot predict the invasion and metastasis of skin squamous cell carcinoma in situ.5.There was a significant positive correlation between eIF4 E and P-4EBP1 positive expression rates in SK and BD tissues,and likewise,there was a significant positive correlation between Ets2 and P-4EBP1 positive expression rates in cSCC tissues. |
