Objective:In this study,the changes of water composition in normal rats with different ages were recorded,and the effects of water composition,including intracellular fluid,on the body water composition of rats with different ages were observed.Methods:1.animals and groupingThe rats of different age were fed with drug-containing feed for 70 days.The initial animals were divided into 8 weeks old group(group 1)and 8 months old group(group 2),And divided into blank groups under the 8-week age group,K1 group,Amorphophallus group was recorded as M1 group,Epimedium group was recorded as Y1 group,Eight-month-old group with blank group K2 group,Group M2,Epimedium group Y2 group.2.Timing measurement of bioelectrical impedance technology Using bioelectrical impedance technique,the body composition of rats was measured under anesthesia,including whole water content,intracellular fluid,extracellμL ar fluid and so on.3.Determination of water content Water content of lung,liver and kidney organs was measured by dry and wet method4.Histomorphology observation of main organs of rats The HE staining method was used to observe the organ tissue of each group of rats and the tissue was fixed for 48 h.Then tissue embedding,slicing,dewaxing,dyeing and other steps were observed and photographed under a positive microscope.5.Water channel protein mRNA and protein expressions The corresponding AQPsmRNA gene amplification levels of lung,liver and kidney were detected by fluorescence quantitative PCR(q PCR),and the distribution of AQPs antigen and antibody binding lung,liver and kidney were investigated by immunohistochemistry.Results:1.General state observation in ratThere was no difference in the color and lustre of each group,no difference in the amount of drinking water in Epimedium group > Dry lotus group > blank group,no difference in the amount of drinking water between age groups;8 weeks old groups were more than 8 months old groups,the same month old group was more Epimedium group,dry lotus group and blank group were not different;from defecation and urine,the color of stool affected by feed from black to dark brown to dark yellow,also softened in turn is blank group > dry lotus group > Epimedium group;Activity: Epimedium group(Y1、Y2)was more active in the same age group,and there was no difference between the two groups.2.Water content detection2.1 Changes in water content in normal ratsIn 0-70 days,the total water content of the whole water content and the total water content of the eight-week-old rats increased with the increase of body weight,but the overall water content decreased,while the overall water quantity and the overall water content of the eight-week-old rats showed a downward trend in August.In the amount of extracellμL ar fluid,8 weeks old and 8 months old showed that the increase of extracellular fluid and extracellular fluid content was not obvious 41 days ago.The intracellular fluid showed a downward trend in both ages.2.2 Whole water content The effect of Mohanlian on 8 weeks rats was not significant,and the decrease of overall water content in 8 months old rats was accelerated.Epimedium had a trend of water consumption in 8 weeks group,slowed down the decrease of water content in 8months old rats,maintained that the total water was relatively flat,and had the effect of replenishing water to a certain extent.2.3 Extracellular fluid Epimedium,as a tonifying medicine,inhibited the increase of extracellular fluid to a certain extent,and the effect of tonif ying yang could help the increase of extracellular fluid.2.4 Intracellar fluidEight-week-old rats had little effect on the intracellular fluid,and the eight-week-old rats had better effect on the intracellular fluid than Herba2.5 Organ water content According to the dry-wet specific gravity of the main organs,there was no significant difference in each group,suggesting that the change of water content in the body may not be affected by organ lesions.HE staining 3.liver and kidney tissue2.6 Lung HE staining HE staining showed that the pμL monary tissue structure was normal,the alveolar septum and alveolar cavity were intact,and the interstitial distribution of lung was uniform2.7 HE liver staining All 8 weeks old rats(Fig.1-10 A、B、C)had compact liver structure and no obvious vacuoles in central vein and tissue.The liver tissue structure of 8 months old rats was still dense,and a small amount of hepatic vacuoles were occasionally seen.2.8 Renal HE staining At 8 weeks old and 8 months old,the renal pathological section of the control group HE showed thin and clear glomerular vascular loop,complete renal tubμL ar structure,clear lumen,complete basement membrane,a small amount of renal stroma,neat arrangement of epithelial cells,no vacuolar cell formation,no inflammatory cell infiltration.The glomerular morphology was normal in the two age groups,but the space was more than that in the normal group.3.Expression of lung,liver and kidney AQPs mRNA and immunohistochemical resμL ts The relationship between water content and aquaporin was found to be positively correlated with lung AQP1、kidney AQP2,negatively correlated with lung AQP4、 liver AQP1\AQP9,and uncertain with kidney5.Conclusion:5.1 Water content in normal rats With the increase of age,the whole water content decreased,the intracellular fluid decreased and the extracellular fluid increased.5.2 Effects of Herba Epimedium on Water Content The effect of Mohanlian on the overall water content of 8 weeks rats was not significant,which was helpful for the increase of extracellular fluid in 8 months old rats.Epimedium had a trend of water consumption in 8 weeks group,slowed down the decrease of water content in 8 months old rats,maintained that the total water was relatively flat,and had the effect of replenishing water to a certain extent.In terms of intracellμL ar fluid,eight-week-old rats had little effect on the intracellular fluid,and eight-week-old rats had better effect on the intracellular fluid.5.3 Whole water content With the increase of age,the morphology of organs will change,and organ lesions will affect the change of water content.According to the results of paraffin sections and HE staining,the pathological changes of lung,liver and kidney,especially lung and liver,were not observed.5.4.The relationship between water content and aquaporin was found to be positively correlated with lung AQP1、kidney AQP2,negatively correlated with lung AQP4、liver AQP1、AQP9 and uncertain with kidney.5.5.The water content results of Epimedium suggest that Yang should not be supplemented in childhood that will consume liquid;in old age,Yang is insufficient,Yin loss and protection can enhance the role of "Yang is Yin conservation" and maintain the stability of water content.
