| Objective:To study the effect of GEO(GEO)on the proliferation,migration and apoptosis of human breast cancer cells MDA-MB-231 and MCF-7 in vitro and in vivo,and to explore the mechanism of apoptosis.Part 1: The effect of ginger essential oil on breast cancer MDA-MB-231 cells in vitro.Methods:MTT assay was used to analysis the effect of different concentrations of GEO on the proliferation of MDA-MB-231 and MCF-7 cells in different time periods(24 h,48 h and 72 h);Cell scratch test was used to analysis GEO effect on the migration of MDA-MB-231 cells;Hoechst 33258 staining,Annexin-FITC /PI staining and flow cytometry analysis,microscope observation were used to analysis the apoptosis rate of MDA-MB-231 cells induced by GEO;Effect of GEO on the expression of Bax and Bcl-2 genes in MDA-MB-231 cells by polymerase chain reaction;Enzyme-linked immunosorbent assay(ELISA)to analysis the effect of GEO on the oxidation reaction of MDA-MB-231 cells.Results:MTT assay and cell scratch tests showed that GEO had a significant inhibitory effect on the proliferation and migration of MDA-MB-231 cells.The MTT assay showed that GEO can effectively inhibit the proliferation of MDA-MB-231 and MCF-7 cells.With the increase of the concentration of GEO and the treated time was prolonged,GEO inhibits the proliferation of MDA-MB-231 and MCF-7 cells,the more obvious the time-concentration dependence was.After observing under an inverted microscope,MDA-MB-231 cells were treated with GEO(25,50,100,1500,200 mg/L)for 48 hours.Compared with the control group,the proliferation rate of MDA-MB-231 and MCF-7 cells increased gradually.The adhesion decreases gradually with the increase of GEO concentration,the cell gap increases more obviously.It can be seen that the cell bodies become round,small and even fall off,and floating cells can be seen.The results of flow cytometry indicate that MDA-MB-231 cells has been intervened by GEO,as the concentration of GEO continues to increase,the apoptosis rate increases significantly.The effect of GEO on the migration of MDA-MB-231 cells was detected by cells scratch test.The results showed that GEO can significantly reduce the migration ability of breast cancer cells compared with the control group(DMSO group);PCR detects the effect of GEO on MDA-MB-231 cells after 48 hours,the ratio of Bcl-2 gene expression to Bax gene expression in the experimental group was significantly lower than that in the control group,and 200 mg/L GEO protein expression changing the most.The results of oxidation index test showed that malondialdehyde activity and lipid peroxide level of cells increased after the treatment of GEO,and catalase content decreased first and then increased with the increase in concentration.Conclusion:Ginger essential oil can inhibit the activity of breast cancer MDA-MB-231 cells in vitro,cause changes in the content of cellular lipid peroxides MDA and LPO,down-regulate Bcl-2 and up-regulate the expression of Bax gene and induce cell apoptosis.Part 2: Effects of ginger essential oil on MCF-7 breast cancer cells and tumors induced by environmental estrogen BPA Methods:Qualitative and quantitative analysis of proteome of MCF-7 cells with bisphenol A induced by ginger oil by itraconazole labeling method.Results:The proteomic analysis of human breast cancer MCF-7 cells indicated that GEO inhibited the proliferation of MCF-7 breast cancer cells induced by bisphenol a,which may be related to SDHB,SDHC,COX-2,SOD 2 and other oxidative phosphorylation pathway proteins.Conclusion:Ginger essential oil can inhibit the growth and proliferation of MCF-7 breast cancer cells induced by environmental estrogen BPA in vitro,inhibit the growth of breast tumors in vivo under the intervention of estrogen,and change the protein composition of cells and tissues and the distribution of intestinal microenvironment.And its mechanism of inhibiting tumors in vivo and in vitro is related to the oxidative phosphorylation pathway. |
