Objective: EBV-associated lymphoma and EBV+LPD are two different types of EBV-related lymphopathys.The infection and expression characteristics of EBV in both lesions need to be further studied.This topic compares the replication,transcription and protein expression of EBV infection in EBV-associated lymphoma and EBV+LPD,in an attempt to reveal the infection characteristics and relevance of EBV in two different lesions to a certain extent,providing a theoretical basis for the accurate diagnosis and treatment of these two diseases.Method:1.Collect 33 EBV(+)lymphoma patients and 5 EBV+LPD patient wax blocks from the Department of Pathology,Shaanxi Provincial People’s Hospital(January 2012-December 2020),and perform EBER in situ hybridization and immunohistochemistry LMP1 and EBNA2 detection on all samples.2.Fluorescence quantitative PCR method was used to detect 33 cases of EBV-associated lymphoma and 5 cases of EBV+LPD patients with peripheral blood EBV-DNA copy number.3.Collect 33 cases of EBV-associated lymphoma and 5 cases of EBV+LPD patients clinical data,and follow up all patients.Result:1.The number of EBER positive signals is statistically different in the EBV+DLBCL group and the HL group(P<0.05),and there is no statistical difference in the expression between the EBV-associated lymphoma group and the EBV+LPD group and the rest of the group(P>0.05).EBER is only expressed in HRS cells in HL,and only in B cells in AITL.2.In patients with EBV-associated lymphoma,the peripheral blood EBV-DNA positive rate is 51.5%(17/33),and the positive rate of peripheral blood EBV-DNA in EBV+LPD is 100%(5/5),and there is a statistical difference between the two disease(P<0.05).Among the subtypes of EBV-associated lymphoma,EBV+DLBCL group and EBV+LPD group(P=0.024),HL group and EBV+LPD group(P=0.026),NK/T cell lymphoma group and EBV+LPD group(P=0.010),peripheral blood EBV-DNA loads and positive rates are also different(P<0.05).3..expressed in HRS large tumor cells.It is only partially expressed in other EBV-associated lymphomas.The positive rate of EBV+DLBCL is 40.0%(2/5),and the positive rate of NK/T cell lymphoma is 33.3.%(4/12),the AITL positive rate is33.3%(2/6),all of AITL are weakly positive and expressed in large HRS-like cells scattered in tumor tissues,is B lymphocytes.All five cases in EBV+LPD were negative.LMP1 expression is significantly correlated with EBER and peripheral blood EBV-DNA copy number(P<0.001).4.EBNA2 is only expressed in 2 cases of EBV-associated lymphomas.5.The number of EBER positive signals,peripheral blood EBV-DNA loads,and LMP1 expression are not significantly related to the clinical characteristics and prognosis of EBV-associated lymphoma patients.Conclusion:1.The peripheral blood of EBV-associated lymphoma patients can detect high EBV-DNA copies.There are differences in peripheral blood EBV-DNA load in EBV-associated lymphoma and EBV+LPD,and the number of EBER positive signals and the distribution of tumor cells also have different characteristics.2.LMP1 is highly expressed in HL,EBNA2 expression prompts are related to immune dysfunction.3.LMP1 expression has a significant correlation with EBER and peripheral blood EBV-DNA copies,and is not significantly related to the clinical characteristics and prognosis of EBV-associated lymphoma patients.
