| Background:Depression is a common chronic psychiatric disease that causes a great burden on society and families.The treatment of depression has become a topic that needs to be studied and solved urgently.xiaoyaosan has the effects of soothing the liver and nourishing blood,invigorating the spleen and removing dampness.After thousands of years of verification,it has achieved satisfactory results in the treatment of depression.At present,the pathogenesis of depression is still not fully understood,and the correlation between changes in the PI3K/AKT signaling pathway and the onset of depression has also been extensively studied.This study further explored the effect and mechanism of xiaoyaosan in the treatment of depression through animal experiments.Objective:This topic uses chronic mild unpredictable stress to replicate the depressed rat model,and evaluates the depressed rat model from the general state of the animal,changes in behavior,and objective indicat ors.Explore the relationship between the changes of PI3K/AKT signaling pathway and depression,and explore the mechanism of xiaoyaosan on depression model rats.Provide experimental basis for clinical treatment of depression with xiaoyaosan.Method:Sixty clean SD rats were randomly divided into:xiaoyaosan group,model group,normal group and fluoxetine group,each with 15 rats.CUMS was used to establish a depression rat model.The rats received drug intervention,and the daily dose of rats in each group was calculated according to their body weight.Normal group: normal feeding without any stimulation.Model group :CUMS model was created,and the corresponding volume of distilled water was administered according to their body weight.xiaoyaosan group:the same method as the model group,and xiaoyaosan was administered daily.Fluoxetine group: the same method as the model group,and fluoxetine was administered daily.During the period,the mental state of each group of rats,the degree of response to external thorns,gavage compliance,the shape,color,and texture of the fur of the rats were observed;the food intake and body weight of the rats were measured;in the open field experiment,the statistics of each group The staying time,total moving distance,standing times,and modification times of the rat’s central area;the sugar water preference experiment was used to determine the sugar water prefe rence index of each group of rats;the hippocampal tissue was taken from the rat anesthetized,and the expression of hippocampal PI3K and AKT m RNA was detected by RT-PCR;Rats were fixed by reperfusion,and the expression of hippocampal PI3K and AKT protein was detected by immunohistochemical method.Result:1.The depression model rat was successfully replicated.Rats have general changes such as discomfort,yellow fur,increased ocular secretions,weight loss,and decreased food intake,as well as depression in the sugar preference experiment and the open field experiment.After the intervention of xiaoyaosan,the depression-like behavior of depression model rats was improved.2.The results of RT-PCR experiments showed that: compared with the blank control group,the expression of PI3K and AKT genes in the hippocampus of the model group was reduced,and the difference was statistically significant(P<0.05);compared with the model group,xiaoyaosan The expression of PI3K and AKT genes increased in the group,and the difference was statistically significant(P<0.05).3.The results of immunohistochemical experiments showed that: compared with the blank control group,the protein expression values of PI3K and AKT in the hippocampus of the model group were reduced,and the difference was statistically significant(P<0.05);compared with the model group Compared with xiaoyaosan group,all indicators were up-regulated,and the difference was statistically significant(P<0.05).Protein expression and gene expression changes are basically the same.Conclution:1.The CUMS method can successfully replicate the rat model of depression with liver depression and spleen deficiency for 6 weeks.2.Xiaoyaosan can not only up-regulate the expression of PI3K and AKT in the hippocampus from the protein level,but also up-regulate the expression of PI3K and AKT in the hippocampus from the gene level.This may be one of the biological basis for the antidepressant effect of xiaoyaosan. |
