| Traumatic Brain Injury(TBI)has the characteristics of complex pathogenesis,and there has been a lack of effective treatment methods in clinical practice.Baicalein and baicalin can promote the recovery of brain injury,while ultrasound can non-invasively inhibit brain edema and nerve cell apoptosis.Therefore,this study investigated the protective effects and mechanisms of baicalein and baicalin combined with ultrasound in the treatment of traumatic brain injury.The Fneeny free fall method was used to establish 105 TBI rat models,which were divided into 7 groups: TBI group,sham operation group,ultrasound group,baicalein group,baicalin group,baicalein combined with ultrasound group,and baicalin combined with ultrasound group.14 days of treatment,and development of a fixation device and method for ultrasound treatment of animal brain injury.Magnetic resonance scanning imaging technology was used to scan rats with 3h,1d,4d,7d,10 d,14d DTI and DKI,and the ADC and MK values of the injury site were measured.The ADC measured by baicalein and baicalin were combined with ultrasound at14 days.The values are(0.735±0.010)and(0.738±0.025),and the MK values are(0.516±0.032)and(0.514±0.029),respectively.The results show that the combined group can promote the repair of the internal structure of the brain tissue.Neural function scores were performed,tissue sections were prepared,and HE and Nissl staining were performed.After 14 days,the number of cells in the combined group was significantly increased,and the morphology and size were basically the same.The Nissl bodies were arranged neatly and the loss rate was significantly lower.Baicalein and Scutellaria baicalensis The loss rates of glycosides combined with ultrasonic Nissl bodies were(28.49±3.52)% and(29.52±4.72)%,respectively,indicating that the combined group can effectively improve the neuronal activity of brain tissue.The concentration of IL-1β and TNF-α in each group was measured by enzyme-linked immunosorbent assay technology.The results showed that the concentration of IL-1β protein in TBI rats after 14 days of treatment with baicalein and baicalin combined with ultrasound was(3.776±0.159)and(3.780±0.160)ng/ml,and the TNF-α protein concentration was(1.427±0.175)and(1.430±0.149)ng/ml respectively.It was found that the two protein concentrations in the combination group were both low,and the results showed that nuclear factor decomposition was inhibited It is a single factor,preventing it from participating in the DNA transcription process,blocking the activation of glial cells,and slowing down the movement of cytoinflammatory factors to the lesion to a certain extent,so that the brain injury site can be repaired.In this study,two methods of traditional Chinese medicine and physical therapy were combined,and three methods of magnetic resonance imaging,tissue sectioning and enzyme-linked immunoassay were used to test the therapeutic effect and mechanism of TBI.Studies have shown that the combination therapy is superior to a single therapy and the combined therapy has a synergistic effect.The mechanism is that the combined therapy may be more effective in inhibiting the inflammatory response of TBI.This study provides a new method for clinical treatment. |
