Preliminary Study On The Application Of Plasma Exosome MiRNA Expression Analysis In Adolescent Idiopathic Scoliosis

Objective:In this study,plasma exosomes from patients with idiopathic scoliosis and normal subjects were extracted and their data were statistically analyzed to study the relationship between plasma exosomes and the formation,occurrence and development of idiopathic scoliosis.To study miRNAs in plasma exosomes and establish expression profiles,and to explore the value of miRNAs in plasma exosomes in the early diagnosis of scoliosis.Methods:Peripheral blood of 15 ml was collected from 6 children with adolescent idiopathic scoliosis and 6 healthy children with matched sex and age.Plasma was centrifuged,and exosomes were extracted by hypercentrifugation method.The extracted exosomes were detected by electron microscopy,and the relevant data were observed and statistically analyzed.In addition,miRNAs in exosomes were extracted and sequenced using Next Seq CN500 sequencing platform SE75 mode to screen the differentially expressed miRNAs.The top 30 miRNAs with the most significant differentially expressed(including 15 up-regulated and 15 down-regulated miRNAs)were analyzed by GO enrichment and KEGG enrichment analysis.Results:(1)In this study,12 cases of exosomes from plasma samples were successfully observed under transmission electron microscopy.It can be observed that the extracted exosomes had typical cup shaped or concave disc shaped structure,which was consistent with the electron microscopy performance of exosomes.Statistical analysis was conducted on the extracted exosome data,and no statistical difference was found.(2)In this study,the total read results of miRNAs sequenced showed that the filtered reads of miRNAs in plasma exosomes of children with idiopathic scoliosis were no different from those of normal children,showing no statistical significance.A total of 2491 miRNAs were detected in the two groups,among which the types of miRNAs in the plasma exosomes of the children group were significantly increased,indicating that the increased miRNAs may have a certain influence on the occurrence and development of scoliosis of the spine.A total of 170 miRNAs were differentially expressed between the two groups after screening,with P < 0.05,which was statistically significant.These included 50 upregulated and 120 downregulated miRNAs,We performed GO enrichment on the top 30 miRNAs with the most obvious differential expression(including 15 up-regulated and 15 down-regulated miRNAs),and the results showed that the target gene functions were concentrated in biological processes such as cellular processes,metabolic processes,biological regulation,and regulation of biological process.KEGG pathway analysis of its miRNA showed that its target gene signaling pathways were enriched in metabolic signaling pathways,PI3K/ Akt signaling pathways,MAPK signaling pathways and other signaling pathways.Conclusion:(1)The peripheral blood exosomes of children with adolescent idiopathic scoliosis were roughly the same as those of normal people.(2)There was no statistical difference in the peripheral blood exosomes concentration between the children with adolescent idiopathic scoliosis and the normal controls.(3)Comparison of miRNAs in the plasma exosomes of children with adolescent idiopathic scoliosis and healthy children showed that a total of 170 differentially expressed miRNAs were screened out,of which 50 were up-regulated and 120 were down-regulated.(4)The target genes predicted by the top 30 miRNAs differentially expressed in the plasma exosomes of children with adolescent idiopathic scoliosis are enriched in multiple signaling pathways,such as the metabolic signaling pathway,PI3K/ Akt signaling pathway,and MAPK signaling pathway,and related signaling pathways may influence the occurrence of adolescent idiopathic scoliosis.KEGG pathway analysis of miRNA showed that its target gene signaling pathways were enriched in metabolic signaling pathways,PI3K/ Akt signaling pathways,MAPK signaling pathways and other signaling pathways.
