| Objective: To detect the relative expression of DDB2 mRNA and MMP9 mRNA in normal colorectal,colorectal adenoma(CRA)and colorectal cancer(CRC),and to explore the possible role and relationship of DDB2 mRNA and MMP9 mRNA in the development of "normal tissue→CRA→CRC",so as to provide useful markers for early diagnosis of CRC and provide corresponding theoretical support for targeted therapy of CRC.Methods: 1.Sample collection: Patients who were treated in the Department of Gastroenterology of the Third Affiliated Hospital of Inner Mongolia Medical University from December 2019 to November 2020 were selected,and the intestinal tissue taken under endoscope was used as experimental samples,the patients were divided into groups according to the corresponding pathological diagnosis.2.The experimental groups were divided into three groups: normal group(n=30),CRA group(n=30)and CRC group(n=30).3.Experimental methods: The relative expression of DDB2 mRNA and MMP9 mRNA within the three groups was measured by q RT-PCR.Results: 1.The expression of DDB2 mRNA in normal group,CRA group and CRC group was 1.181±0.415,1.169±0.476 and 0.754±0.233,the difference was statistically significant(P<0.001).Multiple comparison between groups there was no significant difference between the CRA group and the normal group(P=0.882>0.05).There was a statistically significant difference between the CRC group and the normal group(P<0.0001).The CRA group was significantly different from the CRC group(P<0.001).2.The expression of MMP9 mRNA in normal group,CRA group and CRC group was 1.161±0.418,1.485±0.479 and 1.840±0.348,the difference has statistical significance(P<0.001).Compared with the normal group,the expression level of CRA group was increased,and the difference was statistically significant(P<0.05);the expression in CRC group was significantly higher than that in normal group,the difference was statistically significant(P<0.0001);the expression of CRC group was higher than that of CRA group,and the difference was statistically significant(P<0.01).3.In the normal group,the expression of DDB2 mRNA was negatively correlated with MMP9 mRNA,and the difference was statistically significant(r=-0.455,P<0.05).In the CRA group,the expression of DDB2 mRNA was negatively correlated with MMP9 mRNA and the difference was statistically significant(r=-0.590,P<0.001);and in the CRC group,the expression of DDB2 mRNA was negatively correlated with MMP9 mRNA,and the difference was statistically significant(r=-0.583,P<0.001).Conclusion: 1.There was no significant difference in the expression level of DDB2 mRNA between the control group and the CRA group,the expression level of DDB2 mRNA in the CRC group was lower than that in the CRA group and the normal group,suggesting that DDB2 may play an inhibitory role in the occurrence of CRC.2.The expression level of MMP9 mRNA in the three groups increased gradually,suggesting that MMP9 may play a promoting role in the development process of "normal tissue→CRA→CRC".3.DDB2 and MMP9 are involved in the canceration process of CRA.The detection of DDB2 and MMP9 mRNA expression may provide a new theoretical basis for the early diagnosis and treatment of CRC. |
