PurposeGlioma is a primary malignant tumor of the central nervous system.Differ according to the WHO classification,prognosis is different too,there is also a big difference for this part WHO Ⅱ-Ⅲ level survival outcomes for patients with lower grade glioma(LGG).Accurate diagnosis of malignant degree and prognosis is an important basis for developing adjuvant therapy for patients with glioma after surgery.We discovered recently Fam20 c is a novel secretory kinase,which is involved in the signaling pathway of cell cycle through protein phosphorylation.At present,data mining and analysis of public databases is a hot topic in the field of cancer research.The relationship between Fam20 C gene expression and clinical progression of lower grade glioma was analyzed by using the data from the public database and clinical specimens of our own,and the functional enrichment analysis was performed to understand the gene function and signaling pathway,so as to provide new insights into the pathogenesis of tumor and new ideas for clinical treatment.MethodsWe downloaded the Fam20 C gene expression data and clinical data of LGG from TCGA and CGGA official website,and used R software for statistical analysis and application.The correction between all clinicopathological features and Fam20 C expression was performed by Wilcoxon signed rank test,Survival and Survminer package in R were used to compare the difference of overall Survival(OS)between the high expression group and the low expression group by Kaplan-Meier analysis,and P value was calculated by Log-rank test of Survival contained in R.Receiver operating characteristic(ROC)curves were generated by the Survival Roc package in R.Univariate Cox analysis was used to determine potential survival rate,and multivariate Cox regression proportional risk model was used to compare the effects of Fam20 C expression on overall survival rate and disease-free survival rate after adjusting for the corresponding covariates,and to analyze and identify Fam20 C expression as an independent risk factor for OS in LGG patients.Immunohistochemical analysis and HE staining were carried out on the clinical specimens in our hospital,and the results of the database were further verified through the analysis of clinical data.Finally,gene function enrichment analysis was performed for Fam20 C.ResultFam20C was overexpressed in LGG in TCGA,CGGA and clinical specimens from our hospital.The increased expression of Fam20 C was correlated with clinical progress,and the higher expression of Fam20 C was correlated with the poorer OS significantly.Univariate analysis showed that the high expression of Fam20 C was significantly correlated with OS difference.Multivariate analysis showed that the high expression of Fam20 C in LGG patients was independently associated with a significant decrease in OS.Receiver operating characteristic curve analysis showed that Fam20 C had high diagnostic value.GO and KEGG enrichment analysis showed that co-expressed genes were closely related to the biological process of extracellular matrix remodeling,and the related signaling pathways were mainly concentrated in extracellular matrix receptor interaction,cell adhesion,cell apoptosis,cancer pathway,P53 signaling pathway,Notch signaling pathway and cell cycle signaling pathway.ConclusionIn this study,potential prognostic characteristics of LGG patients were established based on TCGA and CGGA databases and clinical samples from our hospital.This feature can divide LGG patients into two groups effectively and the survival outcomes was significantly different.In addition,we identified a potential signaling pathway of Fam20 C in LGG patients.The overexpression of Fam20 C indicated a poor malignant progression and survival in LGG patients,and revealed significant enrichment of EGM receptor interactions,cell adhesion,and apoptosis at the same time.Fam20 C can be an effective molecular marker for clinical prognosis and a potential molecular markera for the prevention and treatment of LGG.Inhibition of Fam20 C can be a possible therapeutic strategy to prevent progression of LGG.
