Objective:To collect the esophageal squamous cell carcinoma(ESCC)and its adjacent normal tissues of Kazak nationality and Han nationality in Xinjiang,and screen their differentially expressed mRNA by gene chip technology,and analyze their bioinformatics,so as to further understand the occurrence and development mechanism of ESCC.method:In this paper,we analyzed the gene chip expression profile of esophageal squamous cell carcinoma(ESCC)mRNA in Kazakh and Han nationalities in Xinjiang,and compared the results of the two groups by using the Wayne diagram,so as to select the differentially expressed mRNA only in ESCC of Kazakh nationality in Xinjiang,and analyze them by Bioinformatics to find the related pathways;construct the protein interaction network to find the core genes.In addition,Kazakh esophageal squamous cell carcinoma and its adjacent tissues were used for RT-PCR;Kazakh esophageal squamous cell carcinoma,Han esophageal squamous cell carcinoma and its adjacent tissues were used for immunohistochemistry to verify the results of microarray analysis.result:1.2490 differentially expressed mRNA were screened out in Kazakh esophageal squamous cell carcinoma.Of the 2490 mRNA,724 genes were up-regulated and 1748 genes were down regulated in Kazakh ESCC.In the Han population,88 genes were up-regulated and 94 genes were down regulated.2.The intersection of differentially expressed mRNA between Kazakh and Han was obtained by Wayne map,and 707 differentially up-regulated mRNA and 1680 differentially down regulated mRNA were finally screened out,which were only differentially expressed in Kazakh esophageal squamous cell carcinoma.These mRNA are involved in the negative regulation of cell differentiation,cell cycle,cell adhesion,glucose and lipid metabolism and other signaling pathways.Cytoscape software found that MMP1,MMP2,met,Ep CAM,flg and sprr1 a were the top ten core genes in the differentially expressed mRNA of esophageal squamous cell carcinoma of Kazakh nationality.3.The high expression of ppfia1 and FADD in Kazakh esophageal squamous cell carcinoma was detected by PCR.The positive rate of plaur in Kazakh esophageal squamous cell carcinoma was higher than that in Han esophageal squamous cell carcinoma.It is consistent with the chip test results.Conclusion1.The mRNA expression of ESCC was different from that of adjacent normal tissues;the mRNA expression of ESCC in Kazakh was different from that in Han.2.The differentially expressed mRNA is involved in a variety of signaling pathways and biological processes,which lays a foundation for further searching for biomarkers related to the diagnosis,treatment and prognosis of esophageal squamous cell carcinoma.
