| Part I:Analysis of differences in FFA and inflammation levels in stroke.Objective:To investigate the differences of serum FFA level and Inflammation level in high-risk population of stroke,and the aim to provide new ideas for screening and prevention of high-risk population of stroke.Methods:We included 101 at high risk and 85 at control by interviewing questionnaire survey in physical examination center.Blood biochemical test results and physical examination were recorded.We collected serum samples,using enzymatic detecting serum FFA level,using the method of ELISA detecting IL-6 and TNF-α,ICAM-1.The changes of FFAand inflammation levels in high-risk group of stroke were investigated by pairwise comparison.Results:Serum FFA、TNF-α、hs-CRP、NLR was statistically different between the control and the high-risk group(P<0.05),and the high-risk group was greater than the control.In stratified analysis of different genders and ages,hs-CRP has statistically significant difference between high-risk and control(P<0.01),and it was higher in high-risk than control.And serum TNF-αlevel in high-risk group was higher than that in control group(P<0.01).Serum FFA in all 60 year olds and female was higher in high-risk than in control group(P<0.01).Conclusions:Serum FFA、TNF-α、hs-CRP level may be markers of stroke in high-risk group.Part II:Serum fatty acid markers in stroke based on targeted metabolomicsObjective:The aim to investigate the differences of serum metabolites that includ 51 kinds of fatty acids in high-risk stroke,and to explore targets for screening and prevention of high-risk of stroke.Methods:20 cases with AIS were included in the First Hospital of Hebei Medical University from September 2020 to December 2020.In the same period by interviewing questionnaire survey in physical examination center,20 high risk of stroke and 20 healthy controls were included.Basic data and routine blood biochemical values were collected.Using GC-MS detection of51 kinds of fatty acid metabolism in serum.The differences about fatty acids in high-risk stroke were explored by pairwise comparison.By comparing AUC,potential fatty acid markers in high-risk of stroke were screened out.Results:There was no significant differences in age,the history of smoking,blood glucose,TG,TC and PLR among three groups(P>0.05).The linear regression equation(R>0.99),RSD≤10%,PCA(R~2>0.5)and PLS-DA(R~2X>0.05,,Q~2>0.05).51 fatty acids standard substances were determined.The method was stable and feasible for samples detection.The results of quantitative analysis showed that the fatty acids of FC>1,P<0.05,including trans-10-decadenoic acid,trans-9-palmitoleic acid,celery acid and erucic acid were satisfied in the high-risk group.Among the fatty acids with statistical difference in pair comparison,the fatty acids with AUC>0.9 in the high-risk group were screened----cis-9-myristic acid,trans-10-decadecenoic acid and trans-9-palmitoleic acid.In addition,the serum cis-9-myristic acid showed an upward trend(P<0.05),while trans-9-palmitoleic acid,erucic acid,trans-10-decadecenoic acid and celery acid showed a downward trend(P<0.05)in the high-risk stroke.In the high-risk group,erucic acid was positively correlated with HDL-C(r=0.466,P=0.039).Conclusions:1.GC-MS detection of serum fatty acid metabolites in human is stable and feasible.2.Cis-9-myristic acid,trans-9-palmitoleic acid,erucic acid,trans-10-decadecenoic acid and celery acid may be serum markers in high-risk population of stroke.3.The decrease of erucic acid concentration may lead to the decrease of HDL-C.It may increase the risk of stroke. |
