Study On The Formulation Technology,quality Standard And Pharmacodynamics Of Compound Zanthoxylum Armatum DC Chilblain Hydrogel

Objective: To explore the preparation technology of compound Zanthoxylum armatum DC Chilblain hydrogel,establish the qualitative and quantitative analysis method of linalool in compound Zanthoxylum armatum DC Chilblain hydrogel and study its pharmacodynamic effect.Method:: Carbom-940 was used as gel matrix.Through orthogonal experiment,the formability,ductility,uniformity,gloss and stability of the compound Zanthoxylum armatum DC Chilblain hydrogel were taken as the main inspection indexes to optimize the preparation process.Thin layer chromatography(TLC)was used to identify linalool in compound Zanthoxylum armatum DC Chilblain hydrogel with n-hexane: ethyl acetate(4:1)as the developing agent and 10% vanillin-sulfuric acid solution as the color developing agent.HP-5 capillary column(30 m × 0.32 mm × 0.25 μm)was used with nitrogen as carrier gas and the flow rate was 1.0 m L/min.The content of linalool in compound Zanthoxylum armatum DC Chilblain hydrogel was determined by gas chromatography(GC)with the injection volume of 1 μL and the split injection ratio of 10:1.The temperature in the vaporization chamber was320 ℃.The rat model of chilblain was selected as the pharmacodynamic evaluation model to determine the percentage of skin lesions,swelling rate and serum indexes of the rats with chilblain,and to investigate the therapeutic effect of the rats with chilblain through pathological sections.Result: The optimal process of compound Zanthoxylum armatum DC Chilblain hydrogel was camphor 4.5 g,essential oil 7.5 g,cinnamon extract 3 g,carbom-940 1.5 g,propylene glycol 45 m L,glycerol 35 m L,triethanolamine 0.25 m L,ethyl hydroxide 0.1 g,and purified water was added to 150 g at last.The results of TLC showed that the color of each spot was clear,RF value was moderate,and negative without interference.The linear range of linalool was0.4-16 mg/m L,with good precision,stability,reproducibility and recovery.Compared with model control group,the contents of serum indexes TXB2,TXB2/6-Keto-PGF1α and IL-1β decreased and the content of 6-Keto-PGF1α increased in the compound Zanthoxylum armatum DC Chilblain hydrogel group.The results showed that the compound Zanthoxylum armatum DC Chilblain hydrogel group had a certain repair effect on the wound healing of rats with chilblain.Conclusion: The compound Zanthoxylum armatum DC Chilblain hydrogel preparation technology is simple,its linalool qualitative quantitative analysis method is accurate,sensitive and easy to operate,strong specificity and good repeatability,can be used for quality control of compound Zanthoxylum armatum DC Chilblain hydrogel therapeutic effects research shows that compound Zanthoxylum armatum DC Chilblain hydrogel has certain treatment function to the frostbite.Its mechanism may be associated with lower inflammatory factor levels and improve the platelet aggregation function,promote the body’s local circulation.
