| Objective:To evaluate the efficacy and safety of resveratrol alone or combined with ablative fractional CO2 laser system(AFL)in the treatment of skin photoaging.Methods:A split-face,controlled clinical trial was conducted.Thirty-two female subjects were assigned to the test group(TG)or the control group(CG)which was respectively applied test product(resveratrol essence)or control product twice a day for 6 months.Each subject was given AFL treatment(laser side)or no laser treatment(non-laser side)on left or right side of the face randomly.Subjective evaluations by investigators and subjects themselves were conducted after treatment.Noninvasive measurements were conducted at baseline and after treatments,including melanin index(MI),erythema index(EI),and cuticle moisture content.Adverse events(AEs)were evaluated during the study period.Results:1.Subjective evaluation by investigatorsAccording to the investigators’assessments,subjects who received the resveratrol essence achieved improvements of their photoaging signs(roughness,fine wrinkles,uneven pigmentation),compared to pretreatment both the laser side and the non-laser side(P=0.008;P=0.033).While the subjects of control group who used resveratrol-free products did not.On the laser side,the resveratrol products produced a better improvement than the control products at 6 months(P=0.045).2.Subjective evaluation by subjectsOn the laser side:at M3,34%,34%and 25%of the subjects were rated as very satisfied,satisfied and slightly satisfied respectively in TG,compared with 11%,28%and 22%in CG.At M6,50%,25%and 17%of subjects in TG were rated as very satisfied,satisfied and slightly satisfied,compared with 17%,34%and 28%in CG.There was no significant difference between the two groups.3.Noninvasive measurementsFor the inter-group comparison of the laser side,the difference values of MI in TG at M2,M3 and M4 were higher than those in CG(P=0.02;P=0.11;P=0.04).Concerning of the difference value of EI or cuticle moisture content,there was no statistically significant difference between TG and CG.For the inter-group comparison of the non-laser side,the difference values of MI in TG at M3,M4,M5 and M6 were better than those of CG(P=0.003;P=0.004;P=0.005;P=0.007).For the difference value of EI or cuticle moisture content,there was no statistically significant difference between TG and CG.4.Adverse eventsOn the laser side:there were 42%in TG and 22%in CG of the subjects experienced mild sting immediately after using the test and control product,and the pain resolved spontaneously within seconds to one minute.Erythema and mild edema occurred in all the subjects of TG and CG immediately after laser treatment.The erythema and edema respectively subsided within 1 day and 3 days after treatment in TG,in contrast to 3-5 days and 5-7 days after treatment in CG.Mild pigmentation was developed in 3%subjects of TG and 11%subjects of CG,and subsided within half a month and 1-3 months after treatments respectively.On the non-laser side:There were 25%and 16%of the subjects in TG and CG experienced slight sting immediately after using the test and control product,which could be resolved by within a few seconds.No other AEs,such as erythema,edema,pigmentation,occurred in all subjects in the two groups.No other AEs like local skin infection or intolerance occurred in all subjects who finished the study.Conclusion:The resveratrol essence alone can improve photoaging;The resveratrol essence can add an efficacy to the AFL treatment and subside the adverse events such as erythema,mild edema,or mild pigmentation induced by AFL. |
