Study On The Relationship Between The Differential Expression Of CircRNAs And Prognosis Of Acute Myeloid Leukemia

Objective:Circular RNAs(circRNA)play an important role in the development and progression of acute myeloid leukemia(AML)。The purpose of this study was to analyze the relationship between the differential expression of circRNAs and the prognosis of acute myeloid leukemia using the GEO database.Methods:The online database GEO was used to download the samples of AML patients.According to the NCCN guidelines,the samples were divided into two groups:high-risk AML group and low-medium-risk AML group.The circRNA/miRNA/mRNA with differential expression was screened by "EDGER" in R software.The survival analysis of circRNAs differentially expressed in the high-risk AML group and the low-medium-risk AML group was conducted through the Survival package of R software,and the ceRNA network was constructed.The relevant circRNAs were validated in different groups of patients.Results:Based on the circRNA/miRNA/mRNA expression data in the GEO database,27 classical circRNAs(hsa-circ-0001764、hsa-circ-2445023、hsa-circ-1362851、hsa-circ-0001097、hsa-circ-0135651、hsa-circ-6273091、hsa-circ-0034693、hsa-circ-0021550、hsa-circ-1294922、hsa-circ-0035426、hsa-circ-0033902、hsa-circ-0001224、hsa-circ-0006609、hsa-circ-0012742、hsa-circ-0065667、hsa-circ-0001228、hsa-circ-0001664、hsa-circ-0016609、hsa-circ-0001247、hsa-circ-0009224、hsa-circ-1453431、hsa-circ-0005524、hsa-circ-0001255、hsa-circ-0003381、hsa-circ-0083137、hsa-circ-0001749、hsa-circ-0230341)were significantly associated with the prognosis of AML patients,and a cRNA network related to these circRNAs was constructed.In addition,the GO and KEGG pathways of mRNAs differentially expressed between the two were also analyzed.Conclusion:By intervening the expression of these circRNAs(hsa-circ-0001764、hsa-circ-2445023、hsa-circ-1362851、hsa-circ-0001097、hsa-circ-0135651、hsa-circ-6273091、hsa-circ-0034693、hsa-circ-0021550、hsa-circ-1294922、hsa-circ-0035426、hsa-circ-0033902、hsa-circ-0001224、hsa-circ-0006609、hsa-circ-0012742、hsa-circ-0065667、hsa-circ-0001228、hsa-circ-0001664、hsa-circ-0016609、hsa-circ-0001247、hsa-circ-0009224、hsa-circ-1453431、hsa-circ-0005524、hsa-circ-0001255、hsa-circ-0003381、hsa-circ-0083137、hsa-circ-0001749、hsa-circ-0230341),the disease progression of AML patients can be delayed or controlled and the prognosis of AML patients can be improved.
