| ObjectiveThis research consists of three parts.By sorting out and analyzing the prescriptions for clinical treatment of chronic and difficult-to-heal wounds,the rule of medication is finally obtained.This provides a reference for the use of traditional Chinese medicine in the treatment of chronic wounds.Secondly,through animal experiments,verify and explore the impact of core prescriptions on chronic refractory wounds in rats,and preliminarily explore its potential mechanism to promote wound healing.Finally,through the analysis of network pharmacology,the potential mechanism of multi-target and multi-compound regulation of the core prescription on chronic refractory wounds.This provides new ideas for new drug development and in-depth mechanism research.MethodsCollected Chinese medicine prescriptions for treating chronic wounds at the First Affiliated Hospital of Guangzhou University of Traditional Chinese Medicine from January 1,2014 to May 31,2020.These prescriptions were prescribed by Professor Feng Huang.Prescription screening is strictly in accordance with the acceptance criteria.Through statistical frequency,association rules and cluster analysis,the frequency of medication and drug combination of chronic wound prescriptions are analyzed,and the potential core prescriptions are finally obtained.To construct a rat model of chronic refractory wounds through animal experiments,and randomly divide them into a blank group,a model group,and an administration group.The general healing and healing rate of rats in each group were observed,and histological observation was performed by HE staining and Masson staining.Based on network pharmacology methods,including disease differential gene analysis,drug active ingredient screening and target acquisition,GO enrichment and KEGG pathway analysis,predict the core prescriptions for the treatment of chronic refractory wounds,the most core compounds and the most important Biological processes and the most meaningful biological pathways.ResultsA total of 402 clinical prescriptions were included for analysis,and a total of 121 traditional Chinese medicines were obtained.The most commonly used drug is astragalus,and the most commonly used drug pair is white lentil and coix seed.The core prescriptions are: Panax Notoginseng,Salvia,Achyranthes Bidentata,Astragalus,Baiji,Angelica,Angelica,White Lentil,Coix Seed.In animal experiments,samples were taken 15 days after intervention.The healing rate of the drug group was significantly better than that of the model group(P<0.05).Observation of the pathological morphology of the skin tissue found that the drug group had better growth of inflammatory cells,collagen deposition,fibroblasts and epidermis than the model group.Through network pharmacology,there are 157 active ingredients and 229 targets in the core prescription for the treatment of chronic refractory wounds.Among them,the core compound is quercetin.GO enrichment analysis shows that the above targets play an important role in regulating oxidative stress.The most critical regulatory pathway is the AGE-RAGE pathway.ConclusionsThis study analyzed and summarized Professor Feng Huang’s clinical medication rules for chronic wounds.Through experimental verification,the core prescription obtained can promote wound healing.The core compound quercetin,oxidative stress in biological processes and the AGE-RAGE pathway were also discovered in the core prescription.This study provides ideas and theoretical basis for expanding the treatment of chronic wounds with traditional Chinese medicine. |
