| Objective:To investigate whether there are differences in single nucleotide polymorphisms of GABA(rs211014,rs211037,rs121909674),SCN1A gene(rs3812718)and SCN2A gene(rs3943809,rs16850331)in epileptic patients with or without a history of febrile seizures,and to further illustrate whether the common single nucleotide polymorphisms in GABA,SCN1A,SCN2A affect the risk of febrile seizures in patients with epilepsy.Methods:The peripheral blood and medical records of patients with epilepsy were collected,and according to the previous history of febrile seizures,the patients were divided into two groups:febrile seizures group and non-febrile seizures group,and then the patients were further divided into subgroups according to the classification standard of 2017 international anti epileptic alliance,namely focal epilepsy with febrile seizures group,focal epilepsy without febrile seizures group,idiopathic generalized epilepsy with febrile seizures group and idiopathic generalized epilepsy without febrile seizures group.The six sites selected in the sample were detected by Snapshot method,and obtained the genotype and allele information of patients.Then,the patients data were analyzed,firstly,the genotype and allele data of epilepsy with febrile seizures group and epilepsy without febrile seizures group were statistically analyzed,secondly,according to the classification criteria,the data of subgroup focal with and without febrile seizures and idiopathic generalized epilepsy with and without febrile seizures were statistically analyzed,finally,the gene scores of GABA and SCN were calculated by two cross-validation approaches,and the association between GABA,SCN gene scores and the epileptic patients with or without a history of febrile seizures was explored by Logistic regression analysis.Results:Our study collected a total of 106 patients with epilepsy as subjects,including 28 epileptic patients with febrile seizures,78 epileptic patients without febrile seizures,9 idiopathic generalized epilepsy patients with febrile seizures,17 idiopathic generalized epilepsy patients without febrile seizures,11 focal epileptic patients with febrile seizures and 48 focal epileptic patients without febrile seizures.Firstly,the statistical analysis of the epilepsy with and without febrile seizures group was carried out.The statistical results showed that the genotype frequency((P=0.896,P=0.921,P=0.350,P=0.471,P=0.219,P=0.222))and allele genes(P=0.795,P=0.855,P=0.400,P=0.694,P=0.469,P=0.805)of rs211014,rs211037,rs121909674,rs3812718,rs3943809,and rs16850331 were no statistically significant difference between the two groups.Then statistical analysis was carried out between focal epilepsy with and without febrile seizures and idiopathic generalized epilepsy with and without febrile seizures.The analysis results showed that there was no significant difference in genotype frequency(P=0.906,P=1.000,P=1.000,P=1.000,P=0.226,P=0.334)and allele(P=0.746,P=0.767,P=1.000,P=0.835,P=0.595,P=0.183)of the above 6 loci between focal epilepsy with and without febrile seizures,and the same was true in idiopathic generalized epilepsy with and without febrile seizures(genotype frequency:P=0.852,P=0.521,P=0.628,P=0.543,P=1.000,P=0.720;allele:P=0.727,P=0.560,P=0.648,P=0.703,P=0.774,P=0.444).The gene scores of GABA and SCN were not related to the history of febrile seizures in patients with epilepsy.(univariate logistic regression analysis:P=0.333,P=0.454,multivariate logistic regression analysis:P=0.783,P=0.410).Conclusion:We observed that there was no significant difference in allele and genotype frequencies of GABA(rs211014,rs211037,rs121909674),SCN1A(rs3812718)and SCN2A(rs3943809,rs16850331)between epilepsy with febrile seizures and without febrile seizures,focal epilepsy with febrile seizures and without febrile seizures,idiopathic generalized epilepsy with febrile seizures and without febrile seizures.The genetic scores of GABA and SCN were not associated with the history of febrile seizures in epileptic patients.These results suggest that the association between GABA,SCN1A,SCN2A and febrile seizures in patients with epilepsy is not clear,and further studies are needed to clarify whether GABA,SCN1A and SCN2A are genetic susceptible factors of FS in patients with epilepsy. |
