| Objective: To explore the relationship between the expression levels of RNA-133b(micro RNA-133b)and soluble vascular endothelial growth factor receptor 1(sFlt-1)in serum and placenta of pregnant women with preeclampsia and their effects on the occurrence and development of preeclampsia.Methods: Thirty patients with preeclampsia,severe preeclampsia and normal pregnant women admitted to the obstetrics department of Linyi Hospital of Traditional Chinese Medicine from December 2018 to May 2020 were selected as the research subjects,and they were respectively set as the preeclampsia group,severe preeclampsia group and healthy control group.1.The expression of sFlt-1 in serum of each group was detected by enzyme-linked immunosorbent assay,and the expression of miR-133 b in serum of each group was detected by real-time quantitative PCR.2.Western Blot assay was used to detect the expression of sFlt-1 protein in the placental tissues of each group,and real-time quantitative PCR was used to detect the expression of miR-133 b in the placental tissues of each group.Results: 1.Serum sFlt-1 protein expression levels of healthy control group,preeclampsia group and severe preeclampsia group were(173.74±26.61)pg/ml,(252.30±44.01)pg/ml,(305.93±32.89)pg/ml,respectively.Serum sFlt-1 protein expression level in preeclampsia group and severe preeclampsia group was higher than that in healthy control group,the difference was statistically significant(P <0.01).Severe preeclampsia group was higher than preeclampsia group,and the difference was statistically significant(P <0.01).2.The expression of miR133 b in the preeclampsia group and the severe preeclampsia group was(0.61±0.37)and(0.42±0.31),respectively,which were significantly lower than that in the healthy control group(0.95±0.35),with statistical significance(P <0.01).The severe preeclampsia group was significantly lower than the preeclampsia group,and the difference was statistically significant(P <0.01).3.The expression level of sFlt-1 protein in placental tissue of pregnant women in preeclampsia group and severe preeclampsia group was(19831.90±4534.51)and(30632.73±1586.23),respectively,which were significantly higher than that of healthy control group(15237.83±1699.52).The difference was statistically significant(P <0.01).The expression level of severe preeclampsia group was significantly higher than that of preeclampsia group,and the difference was statistically significant(P<0.01).4.The expression level of miR-133 b in the placenta of pregnant women in the preeclampsia group and the severe preeclampsia group was(0.58±0.33)and(0.35±0.17),respectively,significantly lower than that in the healthy control group(1.27±0.46),and the difference was statistically significant(P <0.01).The severe preeclampsia group was significantly lower than the preeclampsia group,and the difference was statistically significant(P<0.01).5.The expression of sFlt-1 protein was negatively correlated with the expression of miR133 b in both serum and placental tissues.Conclusion: 1.The expression of sFlt-1 was up-regulated and the expression of miR-133 b was downregulated in serum and placental tissues of preeclampsia patients,showing a negative correlation between the two.2.The expression of miR-133 b in pregnant women with severe preeclampsia is lower than that in pregnant women with preeclampsia,suggesting that miR133 b may be involved in the occurrence and development of preeclampsia. |
