Objectives:The plant Stellera chamaejasme is a perennial herb belonging to the family Thymelaeaceae,and its root is used in traditional Chinese medicine,where it is known as‘langdu’.The root of S.chamaejasme L.have been taken as a traditional medicine to treat psoriasis,skin ulcers,chronic tracheitis,inflamed lymph nodes,tuberculosis and diarrhea for thousands of years.S.chamaejasme has attracted great attention in the last decades.Previous studies have found that some compounds of S.chamaejasme L.shown significant anti-tumor activity.However,compared with the diverse components of Stellera chamaejasme L.,there still is a vast potential for research on its structure and activity.This study intends to investigate the chemical components and biological activities of S.chamaejasme L.intensively to provide a basis for the development of medicinal resources of S.chamaejasme L.Methods and Results:Thirteen guaiane-type sesquiterpenoids based on[5,7]bicyclic system,stelleranoids A–M(1–13),along with six known analogues,were isolated and identified from the roots of Stellera chamaejasme with chromatographic techniques.Their structures including absolute configurations were determined by HRESIMS,NMR spectroscopic data,as well as X-ray crystallographic analysis.Cytotoxicity test in three cell lines indicated that compound 14 had relatively stronger cytotoxic effect against MKN-45,SKOV3 and Du145 cell lines with IC50 of 9.8,17.4 and 7.3μM,respectively;compounds 3 and 8 displayed moderate cytotoxic effect against MKN-45and Du145 cell lines with IC50 ranged from 14.5 to 18.8μM,comparable to those of the positive control.Meanwhile,in LPS-induced RAW264.7 cells model,compounds19 and 20 displayed more potent inhibitory effect on NO production with IC50 of 29.8and 6.3,respectively,than the positive control(L-NMMA,IC50 of 35.1).
