| The liver is the main place where the human body metabolizes exogenous substances.Harmful factors such as alcohol and drugs can directly or indirectly cause liver damage.Lselenium-methylselenocysteine(SeMSC)is a methylated derivative of selenocysteine.Studies have shown that SeMSC has anti-oxidation and anti-tumor effects,but currently there are only few studies on the liver-protecting effect of SeMSC.The intervention effects of SeMSC on the acute liver injury induced by alcohol and CCl4 were separately studied in this thesis,and the liver-protective mechanism of SeMSC in the two types of liver injury was explored.Male Kunming mice were divided into 5 groups: normal group,model group and SeMSC low-dose group(20 μg/kg),middle-dose group(40 μg/kg),high-dose group(80μg/kg).The biphendate(BP)group(200 mg/kg)was added to the CCl4-induced acute liver injury model as a positive control.After 22 days of continuous intragastric administration,mice were modeled by multiple intragastric administration of alcohol or one intraperitoneal injection of CCl4.After the mice were sacrificed,serum was collected to detect serum biochemical indicators,and liver was collected to detect their antioxidant indicators and lipid peroxidation levels.Pathological sections of liver tissue were used to compare liver morphology,and then determine the expression levels of liver tissue cell apoptosis and inflammation-related factors.The main findings are as follows:SeMSC ameliorated liver damage in mice with acute liver injury,and reduced fat accumulation and lipid peroxidation in the liver of mice.SeMSC reduced the activity of ALT,AST,and ALP in the serum of mice with acute liver injury,and reduced liver edema.The liver damage of the drug group improved,the area of liver necrosis was reduced,and the toxicity of alcohol and CCl4 to the liver was reduced.In addition,SeMSC reduced the serum TG and TC levels in mice with acute liver injury,decreased fat accumulation in the liver tissue of the administration group,and significantly decreased the content of lipid peroxide MDA in the liver.SeMSC reduced the degree of oxidative stress in mice with acute liver injury and improved the antioxidant capacity of mice.Compared with the model group,the administration of SeMSC significantly improved the liver GPX and SOD activity of liverinjured mice,and also had a certain effect on the liver CAT activity,T-AOC and GSH content,and the mice’s antioxidant capacity was enhanced.In summary,SeMSC played an anti-oxidant effect in mice with acute liver injury and enhanced the liver’s antioxidant capacity.It reduces the level of oxidative stress in the liver and inhibits liver fatification,thereby reducing liver tissue damage and improving hepatocyte abnormalities,and ultimately protecting the liver. |
