Objectives To invatigate the expression of V-raf murine sarcoma viral oncogene homolog Bl(BRAF)V600E gene mutation and UDP-glucuronosyltransferase1A1(UGT1A1)in papillary thyroid carcinoma(PTC)and nodular goiter(NG),and explore their correlation to clinical pathological features of papillary thyroid carcinoma.Methods The experimental group pathological specimens of 48 papillary thyroid carcinoma patients who received fine needle aspiration biopsy and surgical treatment were collected from the Affiliated Hospital of North China University of Science and Technology during the two years period from 2019 to 2020.The control group pathological specimens of 40 nodular goiter patients who received surgical treatment were collected in same period.Real-time fluorescence quantitative PCR was used to detect the BRAFV600 E gene mutation in puncture specimens of papillary thyroid carcinoma,and the expression of BRAFV600 E and UGT1A1 in papillary thyroid carcinoma and nodular goiter were examined by immunohistochemical mehod.Results 1 The positive rates of BRAFV600 E and UGT1A1 in PTC were 87.5% and77.1%,which were significantly higher than the positive rates of 12.5% and 32.5% in NG,the difference between the two groups was significant(P<0.05).2 The positive rate of BRAFV600 E mutation in PTC puncture specimens was 83.3%,which was slightly lower than the positive rate of 87.5% of BRAFV600 E protein expression in postoperative specimens.There was no significant difference between the two groups(P>0.05).3 The positive expression of BRAFV600 E in papillary thyroid carcinoma was related to capsular invasion and lymph node metastasis,and the difference were significant(P<0.05).4 In papillary thyroid carcinoma,the positive expression of UGT1A1 was closely associated with single lesions,capsular invasion and lymph node metastasis,the difference were significant(P<0.05).5 Lymph node metastasis of papillary thyroid carcinoma is also related to gender,tumor diameter,and capsular invasion.The age,number of lesions,and combined hashimoto’s thyroiditis have no infulence on lymph node metastasis.Conclusions 1 The positive rates of BRAFV600 E and UGT1A1 in PTC was significantly higher than the positive rates in NG.2 The positive rate of BRAFV600 E mutation in PTC puncture specimens is slightly lower than the positive rate in postoperative specimens,but there was high consistency between the two groups.3 The positive expression of BRAFV600 E in papillary thyroid carcinoma is related to capsular invasion and lymph node metastasis.4 The positive expression of UGT1A1 in papillary thyroid carcinoma is related to single thyroid papillary carcinoma,capsular invasion,lymph node metastasis related.5 Lymph node metastasis of papillary thyroid carcinoma is also related to gender,tumor diameter and capsular invasion.Figure 9;Table 6;Reference 160...
