Objectives The research on the interaction of statins is usually animal experiments and clinical trials at present.It consumed a lot of resources.In order to improve the current lack of information on the clinical evaluation data of the interaction between statins and other drugs,this study used data mining modeling for the first time.The effects of drug interactions are described which is from the perspective of the correlation between statin metabolic enzymes(substrate-inhibitor-inducer)and clinical evaluation.The potential drug interactions are predicted,and new research directions are provided for drug interactions..Methods The clinical evaluation data and related literature of statins and other drug metabolic enzymes and drug interactions were collected,translated and verified from DrugBank,KEGG,DrugGuide2007,Ema,Drugs.com,and PubMed,ScienceDirect,Elsevier,Springer Link,etc.According to the pharmacological effect,the drug data was sorted and organized.And the prediction is made by Apriori algorithm.Finally,in order to predict the interaction between statins-other drugs and enzymes.Results 1 The data of drug metabolic enzymes were collated in 26 categories,involving cytochrome enzyme P450,UDP-glucuronosyltransferase,liver carboxylesterase,serum paraoxonase/lactase family.The calculated metabolic enzyme data were mainly concentrated in cytochrome enzyme P450(124625 data)and UDP-glucuronosyltransferase(42980 data).2 The accuracy range of the Apriori model was 0.69-0.77.The association rule for association analysis of statin interaction metabolic enzymes was 291114,and 98 association network maps of statin metabolic enzymes had been established.3 The Apriori model calculated 47911 association rules for atorvastatin drug interactions.When atorvastatin and amlodipine were used in combination,amlodipine reduces the metabolism of atorvastatin(support 17.76%,confidence 73.68%,lift 2.08).Conclusions As an important predictor of drug interactions,metabolic enzymes establish a prediction model of interactions between statins and other drugs based on data mining.This model is the first to successfully use the Apriori to function the relationship which is the enzymes and the clinical assessment.The prediction accuracy of the mould can reach up to 77.57%,which is suitable for the prediction analysis of the correlation analysis of statin-other drug interaction mediated by metabolic enzymes.Figure 15;Table 18;Reference 123...
