| Objectives To observe the effects of Huazhuo Jiedu Shugan Decoction on seizure grade,seizure latency,open field test and water maze test of epileptic model rats and the Changes of Toll-like receptor4(TLR4),Myeloiddifferentiationfactor88(MyD88)and Interleukin-6(IL-6)expression in hippocampal CA1 region,and to explore the antiepileptic mechanism of Huazhuo Jiedu Shugan Decoction,and to lay the experimental foundation for further study of Huazhuo Jiedu Shugan Decoction.Methods 60 male rats were selected and randomly divided into blank group(n = 10)and model group(n = 50).In the model group,Pentylenetetrazol(PTZ)was injected intraperitoneally once every other day for 14 consecutive times to construct the epileptic model.After the model was successfully lit,the rats in the model group were divided into epilepsy model group,sodium valproate group,high-dose Huazhuo Jiedu Shugan Formula group(Huazhuo high),medium-dose Huazhuo Jiedu Shugan Formula group(Huazhuo medium),and low-dose Huazhuo Jiedu Shugan Formula group(Huazhuo low),with 8 rats in each group.Then,huazhuo jiedu shugan formula rats were given different concentrations of drugs by intragastric administration for 21 days.After gavage,the results of open field experiment and water maze experiment in each group were analyzed statistically.Finally,immunohistochemical staining was used to determine the expression of TLR4,MyD88 and IL-6 in hippocampus CA1 region.Results 1 seizure grade: seizure grade of epilepsy model group was significantly higher than that of hyperturbidization group and sodium valproate group,with statistical significance(P<0.01).2 Latency of seizure: The latency of seizure in the hyperturbidity group was significantly longer than that in the epileptic model group,with statistical significance(P<0.01).3 Open field experiment: the upright frequency of the hyperturbidity group was significantly higher than that of the epileptic model group,with statistical significance(P<0.01).The walking distance in the experimental area was significantly longer in the huazhuo high group and huazhuo medium group than that in the epilepsy model group,with statistical significance(P<0.01).The total walking distance in the turbidity group was longer than that in the epilepsy model group,which was statistically significant(P<0.05).The total walking distance of the turbidity height group was significantly longer than that of the epilepsy model group,with statistical significance(P<0.01).4 Water maze experiment: On the third day of the experiment,the swimming distance of blank group and turbidity height group was less than that of epilepsy model group when looking for the platform,with statistical significance(P<0.05).On day 4 of the experiment,the swimming distance of blank group and turbidity height group was significantly less than that of epilepsy model group,with statistical significance(P <0.01).On the fifth day of the experiment,the residence time in the target quadrant of the high turbidity group and the medium turbidity group was significantly longer than that of the epileptic model group,with statistical significance(P<0.01).5Immunohistochemical results: The expression of TLR4 in the hippocampus of the high turbidization group,the medium turbidization group and the sodium valproate group was significantly lower than that of the epileptic model group,with statistical significance(P<0.01).The expression of TLR4 in hippocampus of low turbidity group was lower than that of epilepsy model group,with statistical significance(P<0.05).The expression of MyD88 in the hippocampus of the hyperturbidity group was significantly lower than that of the epilepsy model group,with statistical significance(P<0.01).The expression of MyD88 in hippocampus of Huazhuo group was lower than that of epilepsy model group,with statistical significance(P<0.05).The expression of MyD88 in the hippocampus of the low turbidity group was similar to that of the epileptic model group without statistical significance(P>0.05).The expression of IL-6 in hippocampus of Huazhuo Jiedu Shugan Formula group and sodium valproate group was lower than that of epilepsy model group,with statistical significance(P<0.01).Conclusions 1 Huazhuo Jiedu Shula Decoction can reduce the seizure grade of rats,prolong the epileptic seizure latency of rats,improve the Learning and memory of rats,and relieve anxiety and tension in rats.2 The mechanism of anti-epileptic effect of Huazhuo Jiedu Shugan Fang may be related to interfering with TLR4-mediated MyD88-dependent pathway,decreasing the expression of TLR4,MyD88 and IL-6 in hippocampus,and inhibiting the inflammatory response in brain.Figure 19;Table 12;Reference 159... |
