Object: In this study,the expression of Bcl-2 and Bax in cervical cancer He La cells treated with different drugs was observed.To investigate the effect of albumin paclitaxel combined with cisplatin on apoptosis of He La cells compared with cisplatin alone,and to provide theoretical evidence for chemotherapy treatment of cervical cancer.Methods: according to the drug treatment plan,the He La cells were divided into three groups: group A was blank control group,group B was cisplatin group,and group C was albumin paclitaxel combined with cisplatin group.The concentration of cisplatin was 10μM and of Albumin paclitaxel was 20 u M.Real time PCR and Western blot were used to detect the m RNA and protein expression of Bcl-2 and Bax in He La cells of each group after 24 hours and 48 hours drug treated,respectively.The apoptosis and the morphological changes of He La cells were detected by TUNEL staining.Results: the results of real-time PCR and Western blot showed that the expression of Bax m RNA and protein in He La cells of group A,B and C were different after 24 hours and 48 hours of treatment(P < 0.05),and the expression of Bax m RNA and protein group B and C were significantly up-regulated compared with that of group A(P < 0.05);While,the m RNA and protein expression of Bcl-2 in group B were significantly lower than those of group A(P < 0.05).And typical morphological changes of apoptotic nuclei between group B and C were detect by TUNEL staining.Conclusion: Albumin paclitaxel combined with cisplatin compared with cisplatin alone,It has better antitumor effect.The mechanism may be through reducing Bcl-2 and increasing the expression of Bax to further promote the apoptosis of tumor cells.
