| Objective:Investigate the anti-tumor effect and mechanisms of baicalin on H22 tumor-bearing mice.Methods : The tumor-bearing mice were dividedinto 5 groups randomly,namely model group,CTX group,baicalin 50mg/kg,baicalin 100mg/kg,baicalin 200mg/kg.Model group were gavaged with NS.CTX(20mg/kg)was given to the CTX group,ip.The three dosage baicalin groups were gavaged with50mg/kg,100mg/kg,200mg/kg baicalin respectively.Mice weight was recorded every three days to adjust the lavage volume.After ten days treatment,mice were sacrificed,and tumor,thymus and spleen were weighed immediately.The organ indexes and the rate of tumor inhibition were calculated and recorded.The expression levels of TGF-β1 and IL-6 in tumor tissues were detected by enzyme-linked immunosorbent assay.The expression levels of stat3,bax,bcl-2,caspase-3 proteins and m RNA in tumor tissues were detected by IHC protocol and real-time quantitative PCR.Results:The tumor weight of baicalin 200mg/kg group and the CTX group decreased significantly and the rate of tumor inhibition was 55.7%(P<0.01).The inhibitory rate of the CTX group was 63.16%(P<0.01).The spleen and thymus indexes of the CTX group were significantly lower than model group.The expression levels of TGF-β1 and IL-6 in tumor tissues in the baicalin and CTX groups were significantly lower than model group.Compared with the model group,treatment of 50,100,200mg/kg baicalin could upregulate bax and caspase-3 proteins and m RNA expression and downregulate bcl-2 and stat3 proteins and m RNA expression in a concentration-dependent manner.Conclusion:Baicalin could inhibit the growth of H22 tumor in KM mice.The anticancer mechanism of baicalin may be related to its ability to decrease expression of IL-6,TGF-β1,bcl-2 and stat3,and upregulate bax and caspase-3 expression. |
