| Objective: Probe into the effect of BMSCs on LPS-induced BV2 microglia inflammatory response,and to further explore the relevant mechanisms affecting the inflammatory response,for the treatment of neuropathy Rational pain provides new ideas.Methods: The well-grown BV2 microglia were induced by LPS to establish a neuroinflammatory response model.BMSCs and BV2 microglia cells were co-cultured with good growth status;they were divided into four groups according to different intervention methods:BV2 cell group(Control team),BV2 cell + LPS(LPS team),BMSCs +BV2 cells(BMSCs team),rat bone marrow mesenchymal stem cells +LPS-induced BV2 cells(LPS + BMSCs team).The expressions of TNF-αand IL-6 in BV-2 cells was perceived by RT-PCR and ELISA;the expression levels of JNK,p-JNK,p38,p-p38,IκB-α and p-IκB-α were detected;IF was used to test the expression levels of p65 protein in each team.Results: In contrast to the Control team,the expression level of(IL-6 and TNF-α)in the LPS team was observably up-regulation(P<0.001);In contrast to the LPS team,the expression level of inflammatory factors(IL-6 and TNF-α)in the LPS+BMSCs team was observably down-regulation(P<0.001).In contrast to the Control team,the fluorescence intensity level of p65 in the LPS team was observably up-regulation(P<0.001);In contrast to the LPS team,the fluorescence intensity level of p65 in the LPS+BMSCs team was down-regulation(P<0.05).In p-IκB-α/IκB-α,contrast to the Control team,the expression level of the LPS team was observably up-regulation(P<0.001).In contrast to the LPS team,the expression level of the LPS+BMSCs team was observably up-regulation(P<0.001).In p-JNK/JNK,contrast to the Control team,the expression level of the LPS team was observably up-regulation(P<0.001).In contrast to the LPS team,the expression level of the LPS+BMSCs team was down-regulation(P<0.05).In p-p38/p38,contrast to the Control team,the expression level of the LPS team was observably up-regulation(P<0.001).In contrast to the LPS team,the expression level of the LPS+BMSCs team was down-regulation(P<0.05).Conclusion: In this study,it was found through in vitro experiments that BMSCs can reduce the secretion of TNF-α and IL-6 in activated microglia,and the mechanism probably was associated with the suppression of MAPK and NF-κB pathways.This finding will help further promote research and development.Clinical drugs for the treatment of NP,providing feasible treatment strategies for NP. |
