Objective: To summarize the medication rules of traditional Chinese medicine in the treatment of diarrhea-predominant irritable bowel syndrome by means of data mining,analyze and obtain the core drug combination,and then use network pharmacology to explore the action mechanism of the core drugs in the treatment of this disease.Methods: 1.Randomized controlled trials on oral traditional Chinese medicine in the treatment of diarrhea-predominant irritable bowel syndrome were obtained by searching CNKI,Wanfang,VIP,and CBM databases,and the search time was set from January2012 to December 2021.Screening according to the inclusion and exclusion criteria,extracting relevant data from the included literature,establishing a Chinese medicine treatment database for diarrhea-predominant irritable bowel syndrome,standardizing the aggregated data,and importing the standardized database into the ancient and modern medical record cloud platform,through statistical analysis,association analysis,cluster analysis,complex network analysis to explore the medication rules of traditional Chinese medicine for IBS-D,and analyze the core traditional Chinese medicine combination.2.Query the chemical components and corresponding protein targets of core Chinese medicines through the Traditional Chinese Medicine System Pharmacology Database and Analysis Platform(TCMSP),and then convert the protein targets into corresponding gene names through the protein database(Uniprot).IBS-D-related disease targets were obtained through Gene Cards and OMIM database search,and common target genes of disease and core drug combinations were obtained by Venny software.The drug-ingredienttarget-disease network diagram was constructed by Cytoscape software,and the main active ingredients were screened out.The intersection targets were imported into the STRING database,and a protein network interaction map was constructed to screen out key target genes.The common targets of diseases and drugs were imported into the Metascape platform for GO function and KEGG pathway enrichment analysis,and the mechanism of action of the core drug combination in the treatment of IBS-D was studied.Results:1.After preliminary search,3892 documents were obtained.After screening,267 documents were finally included,including 22 TCM syndrome types,287 TCM compound prescriptions,and 153 TCM flavors.The most common syndromes mainly include liver stagnation and spleen deficiency,spleen and kidney yang deficiency,spleen deficiency and dampness,spleen and stomach weakness,and spleen and stomach damp-heat syndrome.The top five traditional Chinese medicines in the frequency of use are Atractylodes,licorice,white peony,dried tangerine peel,and Poria,and the high-frequency medicines with a frequency of ≥20 are mainly tonic,qi,and diuresis.The four qi of the drug are mainly warm and flat,and the five flavors are mainly sweet,acrid,and bitter,which mostly belong to the spleen,stomach,and lung meridians.The top three drug pairs in the drug association analysis were Licorice-Atractylodes,Atractylodes-Glycyrrhiza,and Atractylodes-Baishao.At the same time,according to the results of the correlation analysis of drugs and syndrome types,it can be concluded that the main drugs for the treatment of IBS-D are Atractylodes,Baishao,Chenpi,Fangfeng,Bupleurum,etc.Cardamom,Psoraleae,Evodia,Schisandra,etc.;the main traditional Chinese medicines for treating spleen deficiency and dampness syndrome are Atractylodes,licorice,Poria,Codonopsis,Coix seed,Chinese yam,Amomum,and white lentil;four types of drug groups can be obtained through cluster analysis.The first category: woody,Coptis,the second category: Citrus aurantium,dried ginger,nutmeg,Coix seed,Chinese yam,Amomum,the third category:Codonopsis,Poria,Atractylodes,licorice,the fourth category:Bupleurum,dried tangerine peel,white peony root,windproof.Through complex network analysis,it can be known that Atractylodes,licorice,white peony root,dried tangerine peel,tuckahoe,Fangfeng,Codonopsis,Bupleurum are the core traditional Chinese medicine prescriptions for the treatment of IBS-D.2.The network pharmacology analysis showed that the main components of the core drug combination were quercetin,kaempferol,7-methoxy-2-methyl isoflavone,luteolin,etc.;Network interaction map,the screened key target genes include AKT1,IL-6,TNF,TP53,IL1 B,etc.;the signal pathways involved in the regulation include the AGE-RAGE signal pathway,IL-17 signal pathway,and TNF signal pathway in diabetic complications,PI3K-Akt signaling pathway,etc.Conclusion: Through data mining,it is found that the most common syndrome of IBS-D is liver stagnation and spleen deficiency syndrome.When formulating and taking medicines,most of the products are warm in nature and sweet in taste.Most of the medicines enter the spleen meridian.,Baishao,Chenpi,Poria,Fangfeng,Codonopsis,Bupleurum 8 flavors are the core combination of traditional Chinese medicine for the treatment of IBS-D.The core traditional Chinese medicine combination contains a variety of active ingredients,which can regulate diseases with multiple targets and participate in the activation of multiple signaling pathways,thereby regulating gastrointestinal motility,protecting the intestinal mucosal barrier,regulating brain-gut axis disorders,improving visceral hypersensitivity and Intervention of IBS-D by regulating mental and psychological factors.
