Non-small cell lung cancer(non-small cell lung cancer,NSCLC)is the most common type of cancer that causes cancer-related deaths worldwide,with a five-year survival rate of less than 15%.Long non-coding RNA(long non-coding RNA,lncRNA)is a non-coding RNA with a length greater than 200 nucleotide(nt).It can participate in regulating the proliferation,metastasis and metabolism of tumor cells and other complex biological processes by binding proteins,stable RNA,participating in the formation of nuclear scaffolds,and guiding the binding of proteins and DNA.One of the biggest obstacles in the clinical treatment of NSCLC is the problem of drug resistance.Therefore,an in-depth understanding of the molecular mechanisms involved in chemotherapy in NSCLC is of far-reaching significance for treating NSCLC and improving the survival rate of patients.This study found that the expression of KCNMB2-AS1 in NSCLC cell lines is upregulated,which can significantly promote cell proliferation,cell migration and cell cycle of NSCLC cell lines,inhibit cell apoptosis,and can significantly promote NSCLC xenograft tumors at the individual level.In addition,in terms of mechanism,the regulatory network of KCNMB2-AS1,mi R-509-3p,and RPS6KA1 in NSCLC was initially confirmed.Using bioinformatics,mi R-509-3p is predicted to be the target gene of KCNMB2-AS1,RPS6KA1 is predicted to be the target gene of mi R-509-3p,and GO functional analysis shows that mi R-509-3p may be involved in the m TOR signaling pathway in.In order to further explore the mechanism of KCNMB2-AS1 in NSCLC,a dual luciferase reporter experiment was performed to verify the relationship between them,and it was determined that the lncRNA-KCNMB2-AS1/mi R-509-3p/RPS6KA1 axis may be through m TOR signal pathway to influence the occurrence and development of NSCLC.In summary,this study proved the tumor-promoting function of KCNMB2-AS1 in NSCL,which aims to provide a new target for the treatment of NSCLC.
